A method to purify the C1s component of the complement system starting from a mix of anormal sera by sequential chromatographies in DEAE-cellulose, TEAE cellulose and Sephacryl S-200 is described. The presence of C1s in the fractions eluted in the different chromatographic runs is detected by nephelometry. The purity of the C1s obtained is determined by immunoelectrophoresis against an antihuman serum protein serum and a specific antiserum. A good quality antiserum, useful for the quatitation of serum Cls, was produced in rabbits starting from purified C1s
