Fungi have sexual mating types that control the specificity of their reproduction. In Mycosphaerella fijiensis, the sexual reproduction is determined by two groups of sexual mating (MAT1-1-1 and MAT 1-2-1). The aim of this study was the molecular characterization of Cuban isolates of M. fijiensis from the available information of their idiomorph sequences. Nine isolates of M. fijiensis were obtained from necrosed leaves of Grande naine in five Cuban localities (Ciego de Ávila, Santa Clara, Santo Domingo, Remedios and Vueltas). The oligonucleotides MfMAT1-1-1 and MfMAT1-2-1 were designed from the information available of the sequences of the gene ideomorphs MAT1-1-1 and MAT1-2-1 of M. fijiensis. DNA fragments of 307 and 546 base pairs were amplified using the polymerase chain reaction technique (PCR). Five M.fijiensis isolates (CCIBP-Pf34, CCIBP-PF39, Pf57-CCIBP, CCIBP-Pf66, CCIBP-PF80) corresponded with the mating groups MAT1-1-1 and four isolates (CCIBP-Pf54, CCIBP-Pf64, CCIBP-Pf110, CCIBP-Pf83) matched with MAT1-2-1. Both sexual mating types were demonstrated to be present in the Cuban isolates of M.fijiensis, and the heterothallic character of this species was ratified. This result will also facilitate the easily obtaining of the sexual state of M.fijiensis in vitro and in the greenhouse in making use of sexually comp6atible isolates.
