Shoot meristem culture is a potential alternative to the more commonly used methods for in vitro cereal regeneration, as they are less genotype-dependent, and more efficiently established seedbeds can be used as donor material. In the present work, de novo morphogenesis processes were induced by indirect organogenesis in apical meristems from in vitro rice plantlets of Reforma cultivar germinated on a MS basal medium. Shoot meristems were obtained when cutting three 2.0-cm segments from the bottom portion of seedling stem. At callus phase, different concentrations of 2,4-D were trialed in the culture medium, also testing the effect of stem segment position on callus formation. In general, two types of calli were observed: morphogenic and nonmorphogenic ones, which differed in their structure and growth. The highest proportion of morphogenic calli was obtained at the bottommost stem segment, using 2.0 mg/L 2,4-D. These calluses were subcultured for 10 days under light or dark conditions, to study their effect on shoot regeneration. At the regeneration media, macro and microelements from MS and N6 media were evaluated separately. From subcultured calluses in dark regenerated more shoots at the regeneration medium with MS macro and microelements rather than with N6, whereas in subcultured calluses in the light, shoot regeneration efficiency diminished radically in both media tested
