The general steps taken to obtain an in vivo-in vitro correlation are reported in this paper. The advantages of obtaining an A level correlation over those of B and C levels are deeply explained. Details are given about how to establish correlations at each level and, specifically, in the case of A level when the dissolution speed depends or not on the test conditions. The areas of application of the in vivo and in vitro correlation, as well as its introduction in the scale-up process for products of modified release are illustrated. it is concluded that and adequate work with the manufacture variables, the optimization of the dissolution methodolody and the in vivo study are the essential toools to establish a correlations at any level.
