The results of a diagnostic protocol for bovine brucellosis in a buffalo (Bubalus bubalis) dairy farm with reproductive failures are informed. A herd of 80 buffaloes was subjected to a screening with the Rose Bengal (RB) and Brucella agglutination (BMAT) tests, and the results subsequently confirmed by using the 2- mercaptoethanol agglutination test (2-ME), the fluorescence polarization assay (FPA) ) and the molecular diagnosis by PCR of the 16S rDNA region of the genus Brucella. The screening detected 51.25% (41/80) RB reactors and 8.75% positive animals (7/80) on BMAT. The confirmatory techniques (2-ME and FPA) detected 6.25% (5/80) of positive animals. The Kappa index was 0.119 for RB compared with 2-ME , 0806 for BMAT against 2-ME, and 1.0 for FPA against 2-ME. A low agreement for RB to 2-mercaptoethanol and high to excellent for agglutination with phenol and FPA was evident. The PCR detected 100% of the confirmed animals (5/5 animals). Serological techniques such as FPA and direct diagnosis by PCR become valuable tools for animal brucellosis diagnosis without bacterial isolation in high risk areas.
