Introduction: Punica granatum L., (Punicaceae) is one species known in Cuba as granate. Some phytocomponents of the fruit show strong antioxidant activity. This plant is used in the traditional herb medicine to treat parasitic infections, diarrheas and dysentery, asthma, flu, fever, inflammation and other illnesses. In recent years, the biological activity of phenols present in the fruit has aroused the interest of both researchers and the drug industry. Objective: to set a lab-scale process that makes it possible to extract total phenols from the fruit. Methods: a maceration process was used to extract phenols from the fruit and the parameters were optimized through experimental designs. The independent variables were time of extraction, drug/menstruum ratio and alcohol concentration. Results: the three variables had significant influences over the process of extraction of polyphenols. Conclusions: under the set working conditions, the optimal extraction is as follows: time of extraction of 72 hours, drug/menstruum ratio of 1:20 and alcohol concentration of 50 % (v/v).
