D-005, lipid substance from the fruit of Acrocomia crispa (corojo palm), contains a mixture of fatty acids, mainly lauric, oleic, myristic and palmitic and has shown anti-inflammatory and antioxidant effects in rodents. The objective of this work was to determine if D-005 induces changes in the frequency of appearance of abnormal forms in the sperm head and/or causes alterations in the sperm concentration. Male Sprague Dawley rats from an oral subchronic toxicity study (90 days of treatment) were used, randomly distributed into four experimental groups of eight animals each: a control treated only with the vehicle and three administered with D- 005 (200, 500 and 1000 mg/kg). Twenty-four hours after the last administration, each animal was necropsied and the right epididymis was extracted to obtain the samples. The sperm concentration was calculated. For the analysis of the morphology of the sperm head, the classification criterion was followed based on normal and abnormal heads that includes amorphous, banana, without hook and two tails. No deaths occurred and no signs of toxicity were detected. The sperm concentration of the animals treated with D-005 were statistically similar to those of the control group. The averages of normal, total abnormal and different abnormal sperm in the treated groups were similar to those of the control group, indicating the genotoxic safety of D-005 on male germ cells. Oral treatment with D-005 (200, 500 and 1000 mg/kg) during the period of spermatogenesis did not reduce the total number of sperm or increase the frequency of abnormal forms, therefore, this substance under study does not have cytotoxic neither genotoxic potential on male germ cells of rats.
