The effect of three pre-milking stimulation routines with bucerro, oxytocin and manual on milking physiological indicators, milk production and composition, was assessed in 26 primiparous buffaloes between 30 and 60 days of lactation, milked manually, belonging from a herd in Mayabeque province. Each treatment lasted five days, seven days off between them, measuring the time of milk ejection, milking time, milk flow, production and composition in stage I and II, residual and total. It was found that milk ejection time was 87.89±15.99s, 76.69±16.83s and 104.15±25.81s for stimulation with bucerro, oxytocin, and manual, respectively. Milking time was 6.47±1.25min, 6.51±1.19min and 6.94±1.23min for the three methods mentioned above. Average milk flow was 0.70±0.15kg/min, 0.73±0.15kg/min and 0.68±0.18kg/min for the three treatments, respectively. The total milk production was not affected by treatments; residual milk production was 0.17±0.03kg, 0.13±0.02kg and 0.23±0.03kg in the order cited above; fat and protein were not affected by treatments, while lactose content in stage II was significantly lower in the oxytocin treatment. It is concluded that manual stimulation produces more dairy ejection time and residual milk, while milk production and flow were not affected by the treatments used.
