GONZALEZ SANTOS, Jorge Antonio et al. Epididymal sperm cryopreservation at different postmortem times in canine. Rev Salud Anim. [online]. 2013, vol.35, n.2, pp. 137-141. ISSN 0253-570X.

The quality of the spermatozoa obtained from canine epididymis tail before and after being cryopreserved at 3, 24, 48 and 72 hours postmortem was evaluated. The sperm was obtained by orchiectomy of sacrificed canines, which after dissection of the epididymis, the sperm was collected by retrograde flow with the diluent Andersen. The progressive motility, viability and acrosomal integrity were evaluated in the samples obtained before and after being cryopreserved at -196°C in liquid nitrogen. The results showed that the average number of spermatozoa with progressive motility decreased significantly (p<0.05) after 48 hours, when values of 70.45±5.88 and 25.85±4.04, postmortem and post-thawed respectively, were obtained. The live spermatozoa also decreased significantly (p<0.05) to obtain percentages of 92.35±1.78 at 3 hours, to of 79.95±3.48 at 24 hours, to 60.75±6.73 at 48 hours and 58.45±3.37 at 72 hours. Likewise, the averages of live spermatozoa at thawing decreased significantly (p<0.05), where the the percentages of live spermatozoa were 73.30±4.64 at 3hours, 61.75±5.46 at 24 hours, 48.95±5.15 at 48 hours and 29.75±5.8 at 72 hours. Finally, the acrosomal integrity was significantly reduced (p<0.05) with the best results observed until the 24 hours, 78.05±4.57 and 61.95±4.55 for post-thawing and postmortem, respectively. It was concluded that the extraction technique with the diluent used and freezing allows a favorable postmortem recovery of sperm from epididymis tail of dogs.
