In order to morphometrically characterize Bubalus bubalis oocytes, 274 oocyte-cumulus complexes were evaluated. They were extracted by the surgical dissection method of the superficial follicles of 48 ovaries. The oocytes were obtained by in vitro maturation. Their capture was carried out under stereoscopic vision with hair, removing the cumulus cells. The indicators evaluated were: total diameter of oocyte, cytoplasm diameter and thickness of the pellucid zone. Oocytes were matured in TCM 199 with 10% FBS for 24 hours at 37oC. After 24 hours of in vitro maturation, the same measurements under microscopic vision were performed. Data were processed by t-test for mean comparison. The values obtained were: for the overall diameter: 154.61µm vs. 167.14µm, for cytoplasm: 145.21µm vs. 148.46µm and for the pellucid zone: 7.85µm vs. 17.79µm (immature oocytes vs. mature oocytes, respectively). It was evidenced that the overall diameter increase responds primarily to the thickness increase of the pellucid zone (PZ) at 9.94µm. This type of characterization has been carried out for Bubalus bubalis for the first time in Cuba, and it is part of the previous elements in performing biotechnologies related to the reproductive process.
