BACKGROUND: parotiditis is an acute contagious disease that mainly affects children aged 5-14 years and requires rapid diagnostic techniques. OBJECTIVES: a capture ELISA was standardized to detect IgM antibodies to parotiditis virus and this method was compared with hemagglutination inhibition. METHODS: one hundred and seventy seven monosera and 285 serum pairs sent by the different provinces from January 2004 to December 2006, as part of the seroepidemiological surveillance of the parotiditis virus, were studied. They were analyzed by the hemagglutination inhibition technique and used for the system standardization. Accuracy of the test was determined and the cross reactivity to other viral agents was also studied. RESULTS: the system evaluation yielded 98.9 % sensitivity, 98.5 % specificity and 98.7 % concordance; good relation was found in the variation coefficient values intra-assays and inter-assay for all the studied cases. No cross reactivity was observed with other infectious agents. CONCLUSIONS: this system proved to be useful for the early diagnosis of parotiditis, more sensitive and rapid than the hemagglutination inhibition.
