Resumen

TORRES LEYVA, Isabel et al. Evolutionary evaluation of phagocytic function of polymorphonuclear neutrophils. Rev Cubana Hematol Inmunol Hemoter [online]. 2004, vol.20, n.2. ISSN 0864-0289.

The neutrophil is the primary responsible for keeping normal defense levels of the host against invasive microorganisms. With the objective of studying the phagocytic function of neutrophils at the same day of their obtaining and 24 hours later, they were isolated from the peripheral blood of 30 donors from the transfusional medicine department using a Ficox-Telebrix 38 (d=1,077 g/cm3 . Cell viability was determined with tripan blue exclusion technique, which was over 95 % in both days. The standardized technique of the Institute of Hematology and Immunology allowed studying opsophagocytic index. Phagocytosis was detected for times T15 and T60. When applying Student t test to paired samples, no statistically significant difference was observed either in cell viability or in the phagocytic function between neutrophils at the day of isolation and 24 hours after. It was concluded that phagocytic function is present even 24 hours after the neutrophils being isolated and preserved under special conditions (RPMI 1640 10 % fetal bovine serum plus D glucose 2mg/ml). This allows studying in vitro effect of different substances such as plant extracts, animal extracts and even synthetic products on neutrophils

Palabras clave : granulocyte; neutrophil; phagocytic function; opsonophagocytic index.
