Since 1980, it is known that vitamin D3 not only has a regulating function in the homeostasis of calcium and phosphorus, but it also plays an important role in the modulation of the immune response. Its action is exerted by joining its nuclear receptor, and it intervenes by activating transcription factors. The receptor is found in cell of different tissues and of the immune system, such as the dendritic cells, macrophages and T lymphocytes. In the immune system, it produces inhibition of the differentiation and maturation of the dendritic cells, interfering in their capacity for presenting antigens to specific T lymphocytes. It also reduces the transcription of genes encoding interleukin 2, interleukin 12, interferon and tumoral necrosis factor , bringing about an imbalance between the auxiliary Th 1 and Th 2 T lymphocytes. The immunorregulating activity of vitamin D3 may be used as a therapeutical strategy in autoimmune diseases and to enhance graft tolerance
