Resumen

Objectives: This study was carried out during 1994 to 1996 and its objective is to state the results of the monoclonal antibody assessment (MAb) antiantigen of the prostatic tissue; secreted by CMC.GG7 hybridome with different normal and pathological tissues. Design: Due to the fact of obtaining a hybridome secretor of monoclonal antibodies that recognize antigens of prostatic tissue with paraffine embedding in the Immunology and Biologic Center of the Medical University of Camaguey. An experimental design was performed for studying the MAb obtained in a panel of normal and pathological tissues with different origins, all in paraffine embedding. Tissue samples, either normal or pathological, embedded in paraffine were supplied by "Manuel Ascunce Domenech" and "Amalia Simoni" Provincial Hospitals and "Hermanos Ameijeiras Hospital, the National Institute of Oncology and Radiotherapy of Havana city. Immunoperoxidase technique was used. It is commonly carried out in the immunohystochemical laboratories of our country. All tissues had a previous exam with Hematoxylin and Eosin for proving quality and tissue origin. Culture supernatants of the CMC.GG7 hybridome were tenfold concentrated for being used in the trials. Endogen peroxidase was inhibited with 30% Hydrogen Peroxidase, 30 volumes in absolute methanol during 30 minutes at laboratory temperature. Results: In normal and tumoral prostatic tissues, examination was performed in all essayed tissues, specifically in the cytoplasm of epithelial luminal and basal cells of the secretory acini and ducts of the prostatic gland, the higher score was that of the benign prostatic hyperplasia. Reactivity was observed with some of the epithelial tissues used such as lung, liver, kidney. Also, the brain and cerebelum were assessed , it did not show evidence neither with lymphatic tissue (ganglion, spleen) nor with tumour samples of glandular origin. It only recognized epidermoid carcinoma of smooth skin and epidermoid carcinoma of hard palate with tumours of mesenchymatous origin, it only marked in micrometastasis of undifferentiated breast carcinoma in lymphatic ganglion. Conclusions: The CMC.GG7 MAb had a pattern of recognition more limitted to normal and tumoral prostatic tissue (adenocarcinoma, benign prostatic hyperplasia and benign prostatic hyperplasia concomitant with carcinoma). It did not recognize lymphoid tissue which is important for the differential diagnosis with tumours of unknown origin.
