Abstract

High serum concentrations of Lp(a), a lipoprotein similar to LDL, are associated with coronary, cerebrovascular and peripheric affections derived from atherosclerosis. Thrombogenic properties and inhibilatory effects on fribinolytic mechanisms are attributed to this lipoprotein. Different methods have been used for serum quantification of Lp(a), among them, those immunoenzymatic (ELISA) are relevant. In this work, the process of obtainment, purification and characterization of one of the biologic reagents necessary for the mounting of an Immunoenzymatic System ELISA that quantifies Lp(a) in human serum is described. This reagent is a polyclonal hyperimmune serum as source of polyclonal IgG of rabbit human anti-Lp(a). Using commercial human Lp(a) two femenine rabbits were immunized according to immunization scheme proposed; tittles of human anti-Lp(a) antibodies in both animals were obtained. With the use of non-chromatographic methods (salt precipitation and ionic inter change) polyclonal IgG of rabbit human anti-Lp(a) was purified. To this purified IgG was determined the level of purity (SDS-PEGE) and biologic activity (double immunodiffusion. The purified biomolecule showed the level of purity necessary for the purposes in which it shout be used, it also recognizes the antigen used in the immunization.
