Abstract

In this study the method of -70ºC freezing, for the preservation of Bordetella pertussis and Corynebacterium diphtheriae, was evaluated. An adequate quality control, which included purity, viability and stability testing of the main characteristics, was carried out to verify culture maintenance. For this study different formulations were tested. The one with the best results was selected and a study of maintenance of the characteristics was evaluated through time. In order to measure certain parameters, some industrial scale processes were performed, using a 35 L Chemap bio-reactor. The values obtained from lyophilization-preserved strains were taken as a reference. Thus, the search for alternatives and solutions to problems in their preservation was carried out. The results obtained suggest their possible inclusion in the already established Maintenance Program.
