Abstract

We have analyzed different parameters to standardize the molecular weight determination of new vaccine antigens by gel electrophoresis in the Phast System and by a chromatography method. Linearity was obtained by SDS-PAGE using the reference curves with correlation and determination coefficients higher than 0.98. Repeatability was acceptable and there were no differences between the results of several analysts obtained on three different days. This method is exact and robust under the recommended conditions. Column efficiency was evaluated in four chromatographic matrixes (Superosa 12, Sephacryl S-100 y Sepharosa CL- 4B y Sephacryl S-1000). Good results in both systems allowed the evaluation of outer membrane proteins and lipopolysaccharide from Vibrio cholerae, Leptospira ssp., Pseudomona aeruginosa, Shigella ssp.
