Se utilizó una fuente de 60Co PX-g-30 con una tasa de dosis 49,43 Gy/min. La curva de supervivencia celular mostró un DL50 de 150 Gy. La cinética de muerte celular fue lineal con un ajuste superior a 98 %. La inducción de eventos de conversión génica fue significativa respecto al control a partir de 50 Gy. Por el contrario, la reversión génica fue significativa solo a partir de 200 Gy. En general, las frecuencias de eventos de conversión génica fueron superiores a las de reversión, esto sugiere que la radiación gamma induce preferentemente eventos recombinogénicos. Tanto para los eventos de conversión como de reversión génica se obtuvo una dependencia exponencial de la dosis de radiación gamma. <abstract abstract-type="short" xml:lang="en"><p><![CDATA[Radiosensitivity and kinetics of induction of gene conversion and reversion events in Sacharomyces cerevisiae strain D7 to gamma radiation at dose ranges from 100 to 800 Gy and 50 to 300 Gy respectively were studied. A source of 60Co PX-g-30 at a dose rate of 49,43 GY/min was utilized. The cell survival curve showed DL50 of 150 Gy. Cell death kinetics was linear and adjusted over 98 %. The induction of gene conversion events was significant in relation to control from 50 Gy on. However, gene reversion was significant only at 200 Gy. Generally speaking, gene conversion event frequencies were higher than those of reversion, which indicates that gamma radiation preferably induces recombinogenic events. Both the conversion and reversion events showed exponential dependence on gamma radiation dose. <body><![CDATA[<p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-weight:bold'>El  ensayo de genotoxicidad con la cepa D7 de <i style='mso-bidi-font-style:normal'>S.  cerevisiae</i> desarrollado por <i>Zimmerman</i> en 1975, permite medir 3 tipos  de eventos génicos diferentes: entrecruzamiento mitótico, conversión y reversión  génica. Dicho ensayo permite evaluar 2 niveles de expresión del daño en el ADN  (daño primario en el ADN y mutación génica) y su utilidad ha sido probada tanto  para agentes químicos como físicos, incluidas las radiaciones ionizantes.<sup>6</sup>  </span></p>    <p><span style='font-size:11.0pt;mso-bidi-font-size:12.0pt;font-family:"CG Times"'>Considerando  el eventual uso de este ensayo en estudios de mutagénesis y antimutagénesis que  utilicen radiación gamma como mutágeno, el presente trabajo tiene como objetivo  caracterizar la inducción de eventos de conversión y reversión génica en la cepa  D7 de <i style='mso-bidi-font-style:normal'>S. cerevisiae</i> frente a este tipo  de tratamiento.</span><span style='font-size:11.0pt;mso-bidi-font-size: 12.0pt'> <o:p></o:p></span></p><h4><span lang=ES-MX>Métodos</span></h4>    <p class=MsoHeading9>Microorganismo  y medios de cultivo</p>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times"'>Fue utilizada la cepa D7 de  <i style='mso-bidi-font-style:normal'>S. cerevisiae</i> desarrollada por <i>Zimmerman</i><sup>7</sup>  en 1975 de genotipo a/</span><span style='font-size:11.0pt;mso-bidi-font-size: 10.0pt;font-family:Symbol;mso-ascii-font-family:"CG Times";mso-hansi-font-family: "CG Times";mso-char-type:symbol;mso-symbol-font-family:Symbol'><span style='mso-char-type:symbol;mso-symbol-font-family:Symbol'>a</span></span><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"'>, ade2-119/ade2-40,  trp5-112/trp5-27, ile1-92/ile1-92. Esta cepa permite evaluar 3 eventos genéticos:  entrecruzamiento mitótico, conversión y reversión génica. Las células de levaduras  conservadas en el medio descrito por <i>Bronzetti</i><sup>8</sup> en 1995 y con  previo chequeo del genotipo, fueron crecidas a 32 </span><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"; mso-bidi-font-family:Arial'>º</span><span style='font-size:11.0pt;mso-bidi-font-size: 10.0pt;font-family:"CG Times"'>C durante 72 h en medio completo (A) hasta fase  estacionaria de crecimiento, centrifugadas a 4 000 rpm durante 15 min, lavadas  y suspendidas en agua estéril. La suspención celular (4-5 x 10<sup>8 </sup>células/mL)  resultante, fue conservada a 8 </span><span style='font-size:11.0pt;mso-bidi-font-size: 10.0pt;font-family:"CG Times";mso-bidi-font-family:Arial'>º</span><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"'>C hasta  su uso en los ensayos de conversión y reversión.<o:p></o:p></span></p>    <p class=MsoNormal style='text-align:justify'><i><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-weight:bold'>Irradiación</span></i><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"'><o:p></o:p></span></p>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times"'>La irradiación se realizó en  una fuente de <sup>60</sup>Co, modelo PX-</span><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:Symbol;mso-ascii-font-family:"CG Times"; mso-hansi-font-family:"CG Times";mso-char-type:symbol;mso-symbol-font-family: Symbol'><span style='mso-char-type:symbol;mso-symbol-font-family:Symbol'>g</span></span><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"'>-30M.  Las células fueron irradiadas a diferentes dosis dependiendo del tipo de estudio,  a una temperatura de 2 </span><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:Symbol;mso-ascii-font-family:"CG Times"; mso-hansi-font-family:"CG Times";mso-char-type:symbol;mso-symbol-font-family: Symbol'><span style='mso-char-type:symbol;mso-symbol-font-family:Symbol'>±</span></span><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"'> 0,5  </span><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"; mso-bidi-font-family:Arial'>º</span><span style='font-size:11.0pt;mso-bidi-font-size: 10.0pt;font-family:"CG Times"'>C. El valor de la tasa de dosis fue de 49,43 Gy/min,  calculada con el dosímetro Fricke<sup>9</sup> Luego de la irradiación las células  fueron colocadas a 4 </span><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-family:Arial'>º</span><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"'>C hasta  su uso.<o:p></o:p></span></p>    <p class=MsoNormal style='text-align:justify'><i><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-weight:bold'>Radiosensibilidad  celular </span></i><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt; font-family:"CG Times"'><o:p></o:p></span></p>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times"'>El estudio de radiosensibilidad  celular, se desarrolló a partir de un cultivo crecido durante toda la noche en  similares condiciones a las antes descritas. Suspenciones celulares que contenían  entre 6-8 x 10<sup>7</sup> células/mL fueron lavadas y suspendidas en <i>buffer</i>  fosfato (pH 7) y distribuidas en viales de irradiación. El rango de dosis gamma  utilizado estuvo entre 100-800 Gy. Fueron plaqueadas diluciones entre 10<sup>-4</sup>  y 10<sup>-6</sup> para el control y las dosis 100 y 200 Gy, y entre 10<sup>-2</sup>  y 10<sup>-4 </sup>para las restantes dosis. <o:p></o:p></span></p>    <p class=MsoNormal style='text-align:justify'><i><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times"'>Eventos de conversión y reversión  génica</span></i><span style='font-size:11.0pt;mso-bidi-font-size: 10.0pt;font-family:"CG Times"'><o:p></o:p></span></p>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times"'>Las células crecidas hasta fase  estacionaria fueron lavadas, suspendidas en agua estéril y distribuidas en viales  de irradiación en concentraciones de 10<sup>7</sup> células/mL, para el estudio  de reversión y de 10<sup>-5</sup> células/mL para conversión. Las células fueron  irradiadas en un rango de dosis entre 50-300 Gy. Para el cálculo de las frecuencias  de conversión y reversión se procedió según lo indicado por Bronzetti, en 1995.<o:p></o:p></span></p>    ]]></body>
<body><![CDATA[<p class=MsoNormal style='text-align:justify'><i><span lang=ES-TRAD style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"; mso-ansi-language:ES-TRAD;mso-bidi-font-weight:bold'>Análisis estadístico </span></i><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"'><o:p></o:p></span></p>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times"'>Los valores medios de viables  totales, convertantes y revertantes fueron calculados mediante una prueba de Kolmogorov-Smirnov  y posteriormente se comprobó la homogeneidad de varianza de los datos por medio  de una prueba de F-máxima. Los valores medios obtenidos por dosis de radiación,  fueron comparados con los controles no irradiados utilizando una prueba de Dunnet  con una significación de 95 %.<sup>10</sup> El ajuste de los datos experimentales  al modelo teórico fue desarrollado en el programa Microcal Origen-PC, versión  5</span>. </p><h4><span lang=ES-MX>Resultados</span></h4>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-weight:bold'>En  el presente estudio, el porcentaje de supervivencia celular fue considerado como  una medida de la toxicidad de la radiación gamma sobre <i style='mso-bidi-font-style:normal'>S. cerevisiae</i>. En estudio previo<sup>12</sup> se obtuvo un valor de DL<sub>50</sub>  de 400 Gy para la cepa D7 de <i style='mso-bidi-font-style:normal'>S. cereviciae</i>.  Las diferencias encontradas entre ambos estudios pudieran deberse al uso de diferentes  tasas de dosis de radiación gamma. Diferentes autores<sup>13</sup> han encontrado  que la tasa de dosis puede ser un factor determinante en la sensibilidad celular  a este tipo de mutágeno. </span></p>    ]]></body>
<body><![CDATA[<p class=MsoNormal style='text-align:justify'><span lang=ES-TRAD style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"; mso-ansi-language:ES-TRAD;mso-bidi-font-weight:bold'>Con el objetivo de caracterizar  la cinética de muerte celular de la cepa D7 producto de la radiación gamma, se  desarrolló un ajuste a los datos experimentales según uno de los modelos que describe  dicho evento en bacteria.<sup>11</sup> Los resultados mostraron valores de coeficientes  de regresión superiores a 0,98 (tabla 1); esto sugiere que dicho modelo puede  también describir los eventos de muerte celular en levaduras.</span></p>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-weight:bold'>Se  ha planteado que las roturas de doble cadena en la hélice producidas por irradiación  gamma son determinantes en la muerte celular.<sup>14</sup></span><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"; mso-ansi-language:ES-TRAD;mso-bidi-font-weight:bold'> </span><i><span style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"; mso-bidi-font-weight:bold'>Dardalhon</span></i><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-weight:bold'> y  otros,<sup>12</sup> encontraron que las roturas de doble cadena son las responsables  de la muerte celular cuando la cepa D7 de <i style='mso-bidi-font-style:normal'>S.  cerevisiae</i> es tratada con este mutágeno. Dichos autores mostraron que a dosis  de 100 Gy se podía detectar este tipo de daño. Los resultados aquí presentados  están acordes con los obtenidos por estos autores.</span></p>    <p class=MsoBodyText style='text-align:justify'><span lang=ES-TRAD style='font-size:11.0pt;mso-bidi-font-size:10.0pt;font-family:"CG Times"; mso-ansi-language:ES-TRAD;font-weight:normal;mso-bidi-font-weight:bold'>Como se  conoce, el ensayo de conversión génica permite obtener un estimado del nivel de  daño primario producido en el ADN. En este son detectadas únicamente lesiones  potencialmente recombinogénicas, las cuales son susceptibles de ser reparadas.  Por el contrario, la reversión génica es una medida del nivel de mutación fijada  en la célula. Los resultados obtenidos con los ensayos de conversión y reversión  génica sugieren que a dosis de hasta 100 Gy una buena parte de los daños producidos  por la radiación gamma pueden ser reparados, dado que estos no son fijados como  mutación. El hecho de que a dosis menores de 100 Gy, la fracción de supervivencia  (fig. 1) es aún relativamente alta (fracción de supervivencia de 60 %), sostiene  esta hipótesis. </span><span lang=ES-TRAD style='mso-ansi-language:ES-TRAD'><o:p></o:p></span></p>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-weight:bold'>En  general, la frecuencia de convertantes obtenidos en el presente trabajo fue superior  a la de revertantes, lo que sugiere que la radiación gamma induce preferentemente  eventos recombinogénicos. Estos resultados están en correspondencia con los obtenidos  por <i>Hoffmman</i> <i style='mso-bidi-font-style: normal'>y otros</i>.<sup>15-17</sup> No obstante, el ensayo fue lo suficiente  sensible para detectar mutación, es posible que de diverso origen y preferentemente  inducido por lesiones del tipo modificación de bases. Otros autores, han mostrado  que la radiación gamma produce mutaciones génicas en otros tipos de microorganismos  como <i style='mso-bidi-font-style:normal'>Salmonella typhimurium</i><span style='mso-bidi-font-style:italic'>.<sup>18-20</sup></span></span></p>    <p class=MsoNormal style='text-align:justify'><span style='font-size:11.0pt; mso-bidi-font-size:10.0pt;font-family:"CG Times";mso-bidi-font-weight:bold'>El  ajuste de los datos obtenidos a un modelo teórico (fig. 2) evidenció que tanto  los eventos de conversión como de reversión siguen una dependencia exponencial  de la dosis de radiación, con un ajuste entre datos experimentales y modelo teórico  superior a 95 %. La cinética de inducción descrita por estos eventos en <i style='mso-bidi-font-style:normal'>S.  serevisiae</i> es similar a la obtenida por otros autores en <i style='mso-bidi-font-style:normal'>Escherichia  coli</i><span style='mso-bidi-font-style:italic'>,<sup>21</sup></span> cuando  el daño primario producido sobre el ADN se estima de forma indirecta por medio  de genes reporteros de respuesta SOS en bacteria. Both the conversion and reversion events showed exponential  dependence on gamma radiation dose.
