El agente etiológico de esta enfermedad es el virus de la lengua azul (VLA) que se transmite principalmente por vectores hematófagos del género Culicoides y tiene una distribución global. Cuba se encuentra en una zona de circulación endémica del virus y a pesar que desde el año 2007 se reporta a la Organización Mundial de la Sanidad Animal (OMSA) la presencia de bovinos seroreactores, no existen evidencias de manifestaciones clínicas de la enfermedad ni se ha aislado el virus. El objetivo del presente trabajo fue aislar e identificar el virus de la lengua azul a partir de la sangre de un ternero naturalmente infectado de la provincia Mayabeque en 2018. El virus fue aislado en huevos embrionados de pollo y en cultivo celular a partir de la sangre del ternero a los 21 días de edad que resultó positiva a la presencia de virus por un ensayo de reverso transcripción acoplada a la reacción en cadena de la polimerasa anidada (nRT-PCR). Los embriones inoculados mostraron mortalidad embrionaria, disminución de la talla, coloración rojiza y/o edema como indicativo de multiplicación viral. En el cultivo celular se observaron focos de células redondeadas y refráctiles. La identificación del virus se realizó por los ensayos de nRT-PCR, un Kit comercial de RT-PCR en tiempo real (rRT-PCR) y rRT-PCR basado en SYBR Green-I para la detección especifica del VLA.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[ABSTRACT Bluetongue (BT) is a viral disease that affects domestic and wild ruminants. The etiological agent of this disease is bluetongue virus (BTV) which is mostly transmitted by blood-borne vectors of the Culicoides genus and has a global distribution. Cuba is located in a zone of endemic circulation of the virus. Although the presence of bovine reactors has been reported to the World Organization for Animal Health (WOAH) since 2007, there is no evidence of clinical manifestations of the disease and the virus has not been isolated. This work aimed to isolate and identify bluetongue virus from the blood of a naturally infected calf from Mayabeque province in 2018. The virus was isolated in chicken embryonated eggs and in cell culture from the blood of the calf at 21 days of age, which tested positive for the presence of virus by a nested reverse transcription polymerase chain reaction assay (nRT-PCR). The inoculated embryos showed embryonic mortality, decreased size, reddish coloration and/or edema as indicative of viral multiplication. In cell culture, rounded and refractile cell foci were observed. Virus identification was performed by nRT-PCR assays, a commercial real-time RT-PCR kit (rRT-PCR) and rRT-PCR based on SYBR Green-I for specific detection of BTV.]]></p></abstract>
