Esto fue confirmado por RT-PCR.<sup>2</sup>      En contraste, <span class=SpellE>hardjoprajitno</span> tiene una región <span class=SpellE>intergénica</span> <i>J14 - J15 </i>de longitud similar a su homólogo      en <span class=SpellE>hardjobovis</span> pero sin <span class=SpellE>transposasas</span>.      Análisis de <span class=SpellE>transcripción</span> por RT-PCR revelaron una      sola unidad de <span class=SpellE>transcripción</span> en <span class=SpellE>hardjoprajitno</span>      desde <i>J1 </i>hasta <i>J31.</i> Análisis más detallados mediante extensión      del iniciador (PE) demostraron un sitio de inicio de la <span class=SpellE>transcripción</span>      asociado con secuencias similares a promotores s<sup>70</sup> de <i>E. coli      </i>arriba de <i>J15 </i>en <span class=SpellE>hardjoprajitno</span>.<sup>1,      8</sup></span></p>       <p class=MsoNormal><span lang=ES>Para confirmar el papel de esas secuencias      como promotores para <i>Leptospira</i>, se construyó un vector sin promotor      para kanamicina, <span class=SpellE>pGL4W74</span> utilizando como base el      vector <span class=SpellE>pGKLep4</span>.<sup>7</sup> Un fragmento doble <span class=SpellE><i>Sau</i>3AI</span> de 480 bp que contenía el supuesto promotor      arriba de <i>J15,</i> fue clonado en <span class=SpellE>pGL4W74</span>. La      <i>E. coli</i> que contenía el <span class=SpellE>plásmido</span> recombinante,      <span class=SpellE>pGL4W15</span>-96, fue cultivada en caldo LB con 0, 5, 10, 25, 50,      100 y 250 <span class=SpellE>mg</span>/<span class=SpellE>mL</span> de kanamicina      en agitación a temperaturas de 25, 30, 37 y 42 <span class=SpellE>°C</span>.      Se midió la OD<sub>600</sub> por triplicado a las 0, 1, 2, 3 y 24 h y cada      ensayo fue también repetido por triplicado.</span></p>       <p class=MsoNormal><span lang=ES>El <span class=SpellE>constructo</span>, <span class=SpellE>pGL4W15</span>-96, restauró la <span class=SpellE>transcripción</span>      del gen de resistencia a kanamicina, lo que resultó en colonias <span class=SpellE>Ka<sup>R</sup></span> de <i>E. coli </i>y <i>Leptospira <span class=SpellE>biflexa</span></i><span style='mso-bidi-font-style:italic'>,<i> </i></span>confirmando      así, la función del inserto como promotor para ambos organismos. <i>E. coli      </i>recombinantes mostraron una <span class=SpellE>absorvancia</span> de hasta      1,7 en todas las concentraciones de kanamicina a 30 y 37 <span class=SpellE>°C</span>;      pero <span class=SpellE>absorvancias</span> tan bajas como 0 a 25 y 42 <span class=SpellE>°C</span>      en concentraciones de 50 <span class=SpellE>mg</span>/<span class=SpellE>mL</span>      de kanamicina.</span></p>       <p class=MsoNormal><span lang=ES>La <span class=SpellE>transcripción</span>      de los loci <i>rfb</i> de ambos subtipos de la serovariedad <span class=SpellE>hardjo</span>      en 2 unidades está conservada a pesar de las diferencias estructurales en      sus regiones <span class=SpellE>intergénicas</span> <i>orf14-orf15</i>. La      <span class=SpellE>transcripción</span> regulada por el fragmento de 480 bp de esta      región <span class=SpellE>intergénica</span> en <span class=SpellE>hardjoprajitno</span>,      es dependiente de temperatura, siendo activa a 30 y 37 <span class=SpellE>°C</span>,      pero no activa a 25 ni 42 <span class=SpellE>°C</span>.</span></p>       ]]></body>
