Ambas cepas no mostraron diferencias significativas en cuanto a sus características morfológicas. Los mosquitos pertenecientes a Santiago de Cuba Techo presentaronn los mismos parches que los de Santiago de Cuba. La resistencia a los insecticidas organofosforados es muy similar en ambas poblaciones, sin embargo, difieren en la alta resistencia al piretroide deltametrina en Santiago de Cuba Techo en comparación con Santiago de Cuba. Desde el punto de vista bioquímico y mediante el uso del sinergista DEF se demostró que las esterasas están asociadas con la alta resistencia a clorpirifos en ambas cepas, no resultando así las MFO, lo cual se demostró con el sinergista piperonil butóxido. Sin embargo, la enzima GST parece ser la responsable de la alta resistencia detectada a deltametrina en Santiago de Cuba Techo por el valor elevado de frecuencia de ese gen en esta cepa. Se utilizó la técnica del ADN polimórfico amplificado a la azar para observar la variabilidad genética entre las 2 poblaciones; los resultados revelaron que existió polimorfismo genético entre las poblaciones en estudio, lo cual pudiera tener una implicación en la ecología y epidemiología del vector.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Comparison of 2 populations of Aedes aegypti mosquitoes from Santiago de Cuba with different rest conduct]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Two populations of Aedes aegypti that were collected in Santiago de Cuba during the epidemics of 1971 were separated for having different rest habits, some of them rested naturally on the walls up to 1 m high (Santiago de Cuba strain) and the others were found resting in the roofs of the houses (Santiago de Cuba Techo strain). These strains did not show significant differences as regards their morphological characteristics. The mosquitoes corresponding to Santiago de Cuba Techo strain presented the same patches that those of Santiago de Cuba. The resistance to organophosphate insecticides is very similar in both populations; however, the Santiago de Cuba Techo strain shows a higher resistance to pyrethroid deltamethrin than the Santiago de Cuba strain. From the biochemical point of view and by using the DEF synergist, it was proved that esterases are associated with the high resistance to clorpirifos in both strains. It was not so with the MFO, which was demonstrated by means of the piperomyl butoxide sinergist. Nevertheless, the GST enzyme seems to be responsible for the high resistance to deltamethrin detected in the Santiago de Cuba Techo strain due to the elevated frequency value of that gene in this strain. The random amplified polymorphic DNA technique was used to observe the genetic variability between the 2 populations. The results revealed that there was genetic polymorphism between the populations under study, which could have an impact on the ecology and epidemiology of the vector.]]></p></abstract>

Este vector desempeña un papel muy importante  en la transmisión de numerosas <span class=spelle>arbovirosis</span>.<sup>1</sup>  </span>Un método muy útil como apoyo para la supresión de epidemias de dengue  y FHD es el empleo de insecticidas en ultra bajo volumen (UBV<span class=grame>),</span><sup>2</sup>  lo cual en aplicaciones repetidas puede causar 100 % en el control de poblaciones  de mosquitos adultos.<sup>3-5 </sup>La efectividad de este tratamiento puede disminuir  por varias razones, entre las que se encuentra la conducta de reposo de las poblaciones  de <i style='mso-bidi-font-style:normal'><span lang=ES-MX style='mso-ansi-language:ES-MX;mso-fareast-language:EN-US'>Aedes <span class=spelle>aegypti</span>.</span></i><sup><span lang=ES-MX style='mso-ansi-language: ES-MX;mso-fareast-language:EN-US'>3</span></sup><span lang=ES-MX> </span></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><span lang=ES-MX style='mso-ansi-language:ES-MX;mso-fareast-language:EN-US'>Recientemente,  el uso de técnicas moleculares que detectan variación en el nivel de ADN ha permitido  el análisis de marcadores genéticos con alto grado de polimorfismo en regiones  diferentes del genoma.<sup>6</sup><sub> </sub>Fragmentos de ADN obtenidos por  la técnica del RAPD han sido empleados para estudios de variabilidad genética  entre las poblaciones de <i style='mso-bidi-font-style: normal'>Aedes <span class=spelle>aegypti</span></i>.<sup>7,8</sup> </span></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><span lang=ES-MX style='mso-ansi-language:ES-MX;mso-fareast-language:EN-US'>En 1997  ocurre la tercera epidemia de dengue en la ciudad de Santiago de Cuba, en la cual  se reportaron un total de 3 012 casos clínicos, con 205 casos de FHD y 12 muertes,  producidas por el <span class=spelle>serotipo</span> </span>2.<sup>9</sup> En  esta epidemia se <span lang=ES-MX style='mso-ansi-language:ES-MX'>colectaron mosquitos  <i style='mso-bidi-font-style:normal'>Aedes <span class=spelle>aegypti</span></i>  que se encontraban reposando indistintamente, unos reposaban de forma normal en  las paredes hasta 1 m de altura y otros se encontraron reposando en los techos  de las viviendas, de ahí se criaron 2 poblaciones de </span><i style='mso-bidi-font-style:normal'><span lang=ES-MX style='mso-ansi-language: ES-MX;mso-fareast-language:EN-US'>Aedes <span class=spelle>aegypti</span></span></i><span lang=ES-MX style='mso-ansi-language:ES-MX'> separadas en el insectario, con el  objetivo de determinar si estas diferencias en su comportamiento de reposo estaban  asociadas con patrones de resistencia a insecticidas o si existía alguna característica  genética que los diferenciara, lo cual pudiera tener una implicación en la ecología  y epidemiología del vector. En este trabajo se analizaron las características  morfológicas, bioquímicas y moleculares de ambas poblaciones de <i style='mso-bidi-font-style:normal'>Aedes  <span class=spelle>aegypti</span></i> de Santiago de Cuba.</span></p><h4>Métodos</h4>    ]]></body>
<body><![CDATA[<p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><i style='mso-bidi-font-style:normal'>Cepas a utilizar</i></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Para  el trabajo se utilizaron 3 cepas:</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Cepa  ROCKEFELLER<b style='mso-bidi-font-weight:normal'>: </b>cepa susceptible de referencia,  suministrada por el Centro de Control de Enfermedades de San Juan, Puerto Rico.  </p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><span lang=ES-MX style='mso-ansi-language:ES-MX'>Cepa Santiago de Cuba:<b style='mso-bidi-font-weight:normal'> </b>colectada en 1997 en la misma ciudad  y mantenida en condiciones de insectario en el Instituto de Medicina Tropical  &#8220;Pedro <span class=spelle>Kourí</span>&#8221;. Los mosquitos de esta cepa  reposan en las paredes de las casas.</span></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><span lang=ES-MX style='mso-ansi-language:ES-MX'>Cepa Santiago de Cuba Techo:<b style='mso-bidi-font-weight:normal'> </b>colectada en 1997 en la misma ciudad  y mantenida en condiciones de insectario en el Instituto de Medicina Tropical  &#8220;Pedro <span class=spelle>Kourí</span>&#8221;. Los mosquitos de esta cepa  reposan en el techo de las casas.</span></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><i style='mso-bidi-font-style:normal'>I<span style='mso-no-proof:yes'>nsecticidas  grado técnico utilizados</span> </i></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><span class=spelle>Organofosforados</span>: <span class=spelle>malatión</span>, <span class=spelle>clorpirifos</span>, <span class=spelle>metil</span>-<span class=spelle>pirimifos</span>, <span class=spelle>temefos</span> y <span class=spelle>fentión</span>. </p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><span class=spelle>Piretroides</span>: <span class=spelle>deltametrina</span>, <span class=spelle>cipermetrina</span><b style='mso-bidi-font-weight:normal'><i style='mso-bidi-font-style:normal'>.</i></b></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><i style='mso-bidi-font-style:normal'><span lang=ES-MX style='mso-bidi-font-size: 11.0pt;mso-ansi-language:ES-MX'>Análisis morfológico </span></i></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Para  el estudio morfológico fueron utilizados 20 mosquitos adultos de cada cepa y se  analizaron la distribución, posición, tamaño y la intensidad del color de los  parches de cada uno de los individuos<span lang=ES-MX style='mso-ansi-language: ES-MX'>. </span></p>    ]]></body>
<body><![CDATA[<p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><span class=spelle><i style='mso-bidi-font-style:normal'><span style='mso-bidi-font-size: 11.0pt'>Bioensayos</span></i></span><i style='mso-bidi-font-style:normal'><span style='mso-bidi-font-size:11.0pt'> de larvas</span></i></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>En  los <span class=spelle>bioensayos</span> de la OMS<span class=grame>,<sup>10</sup></span><sup>  </sup>se realizaron 5 réplicas de cada concentración del insecticida. (20 larvas  por réplica) registrándose entre 2 y 98 % de mortalidad. Todas las soluciones  se ajustaron a un volumen final de 1 mL con acetona. Esta concentración de acetona  no causó mortalidad en los controles. </p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>La  lectura de las mortalidades se realizó a las 24 h, hallándose la CL<sub>50</sub>  y la CL<sub>90</sub> mediante el programa <span class=spelle>Probit</span>- logaritmo.<sup>11</sup></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>La  acción de 2 <span class=spelle>sinergistas</span>, <span class=spelle>S</span><span class=grame>,S,S</span> <span class=spelle>tributil</span> <span class=spelle>fosforotritioato</span>  (DEF) y <span class=spelle>piperonil</span> <span class=spelle>butóxido</span>  (PB) fueron investigados exponiendo las larvas de cuarto estadio a 0,008 <span class=spelle>mg</span>/L de DEF o 5 <span class=spelle>mg</span>/L de PB durante  4 h previo a la adición de la solución del insecticida y se determinó la mortalidad  después de 24 h de exposición, hallándose las concentraciones letales 50 y 90  a través del programa <span class=spelle>Probit</span>-logaritmo.11 No existió  mortalidad a estas concentraciones del <span class=spelle>sinergista</span> solo.</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Ensayos  bioquímicos: Se determinó la actividad de <span class=spelle>esterasas</span>  en larvas de tercer estadio tardío o cuarto temprano, de acuerdo con el método  estandarizado para <i style='mso-bidi-font-style:normal'>Aedes <span class=spelle>aegypti</span>.</i><sup>12 </sup>Se homogeneizó cada larva en 200  &#956;L de <i style='mso-bidi-font-style:normal'>buffer</i> fosfato 0,01 M, <span class=spelle>pH</span> 7,5. En una placa de <span class=spelle>microtitulación</span>  de ELISA, a 20 &#956;L del <span class=spelle>homogenato</span> se le añadió 200  &#956;L del sustrato (0,7-<span class=spelle>mM</span> de <span style='font-family: Symbol;mso-ascii-font-family:"Times New Roman";mso-hansi-font-family:"Times New Roman"; mso-char-type:symbol;mso-symbol-font-family:Symbol'><span style='mso-char-type: symbol;mso-symbol-font-family:Symbol'>b</span></span>-<span class=spelle>naftil</span>  acetato). Después de dejar transcurrir la reacción por 10 min. Se le añadió 40  &#956;L de <span class=spelle><i style='mso-bidi-font-style:normal'>fast</i></span><i style='mso-bidi-font-style:normal'> blue </i>y se leyó la densidad óptica (DO)  a 570 <span class=spelle>nm</span> en lector de placas de ELISA <span class=spelle>Labsystems</span> <span class=spelle>iMS</span>, manufacturado en  Finlandia.</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>La  actividad de <span class=spelle>glutatión</span>-S-<span class=spelle>transferasa</span>  (GST) fue determinada de acuerdo con el método de <span class=spelle><i>Booth</i></span>  y otros<span class=grame>,<sup>13</sup></span> y modificado para <i style='mso-bidi-font-style:normal'>Aedes <span class=spelle>aegypti</span>.</i><sup>12  </sup>A 20 &#956;L de cada <span class=spelle>homogenato</span> de larva se le  añadió 250 &#956;L de una mezcla de reacción de 1-cloro-2,4 <span class=spelle>dinitrobenceno</span>  50 <span class=spelle>mM</span>; y de <span class=spelle>glutatión</span> reducido  20 <span class=spelle>mM.</span> Se dejó transcurrir la reacción por 3 <span class=spelle>min</span>  y se leyó la DO a 340 <span class=spelle>nm</span>. Se calculó la actividad específica  de GST, después de calcular la concentración de proteínas en cada <span class=spelle>homogenato</span>  y se expresó como &#956;mol/ <span class=spelle>mg.min</span>. </p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Se  determinó la actividad de la <span class=spelle>acetilcolinesterasa</span> (<span class=spelle>AchE</span>) normal e inhibida con <span class=spelle>propoxur</span>  según el método de <i>Rodríguez</i> y otros.<sup>14 </sup>La preparación de las  muestras es similar a la descrita anteriormente, con la diferencia de que las  muestras se homogeneizaron en 50 &#956;L de <i style='mso-bidi-font-style:normal'>buffer</i>  fosfato, que contenía tritón X-100 1 %. Se prepararon además 2 placas, una para  la determinación de la actividad <span class=spelle>AchE</span> normal y otra  para la actividad <span class=spelle>AchE</span> inhibida con <span class=spelle>propoxur</span>. En ambas placas se adicionaron 20 <span style='font-family:Symbol;mso-ascii-font-family:"Times New Roman";mso-hansi-font-family: "Times New Roman";mso-char-type:symbol;mso-symbol-font-family:Symbol'><span style='mso-char-type:symbol;mso-symbol-font-family:Symbol'>m</span></span>Lde  DTNB (ácido 5-5&#8217;ditiobis 2 <span class=spelle>nitrobenzoico</span>) 0,07  M, <span class=spelle>pH</span> 7,5 y 20 &#956;L de <span class=spelle>acetiltiocolina</span>  yodada (<span class=spelle>Achi</span>) 0,06 M. En la placa donde se midió la  actividad <span class=spelle>Ache</span> inhibida se adicionaron además 10 &#956;L  de <span class=spelle>propoxur</span> 0,025 M. Después de preparadas estas 2 placas  se adicionó en cada pocillo de ambas 20 &#956;L del homogeneizado de las larvas.  La reacción se dejó transcurrir durante 30 <span class=spelle>min</span> y se  leyó la DO a 405 <span class=spelle>nm</span>.</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Un  estimado de la frecuencia de los mecanismos de <span class=spelle>esterasas</span>,  GST y <span class=spelle>AchE</span> fue calculado a partir del número de individuos  susceptibles para cada ensayo, asumiendo que la población se encuentra en equilibrio  de <span class=spelle>Hardy</span>-<span class=spelle>Weinberg</span>.</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Se  realizó electroforesis en <span class=spelle>gel</span> de <span class=spelle>poliacrilamida</span>  (10 %). En tubos <span class=spelle>Eppendorf</span> (1,5 mL), se adicionaron  10 &#956;L de muestra más 10 &#956;L del indicador <span class=spelle>Xilene</span>  <span class=spelle>cianol</span> (0,02 % en sacarosa 15 %). Se aplicaron 20 &#956;L  de esta mezcla en el <span class=spelle>gel</span> y se realizó la corrida a 150  V, durante 45 min. Para la tinción de las bandas de <span class=spelle>esterasas</span>,  se sumergieron los <span class=spelle>geles</span> en 50 mL de <i style='mso-bidi-font-style:normal'>buffer</i> fosfato (0,1 M) que contenían 4  mL de cada uno de los sustratos inespecíficos de las <span class=spelle>esterasas</span>  (<span style='font-family:Symbol;mso-ascii-font-family:"Times New Roman"; mso-hansi-font-family:"Times New Roman";mso-char-type:symbol;mso-symbol-font-family: Symbol'><span style='mso-char-type:symbol;mso-symbol-font-family:Symbol'>a</span></span>  y <span style='font-family:Symbol;mso-ascii-font-family:"Times New Roman"; mso-hansi-font-family:"Times New Roman";mso-char-type:symbol;mso-symbol-font-family: Symbol'><span style='mso-char-type:symbol;mso-symbol-font-family:Symbol'>b</span></span>-<span class=spelle>naftilacetato</span>). Después se añadieron 0,5 g del colorante <span class=spelle><i style='mso-bidi-font-style:normal'>Fast</i></span><i style='mso-bidi-font-style:normal'> blue</i> RR, disuelto previamente en agua  destilada y SDS (sodio <span class=spelle>duodecil</span> sulfato) 5 %. Para fijar  la coloración de las bandas se sumergió el <span class=spelle>gel</span> en una  solución de ácido acético 10 %. A cada una de las bandas se le determinó la movilidad  relativa.</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><i style='mso-bidi-font-style:normal'><span lang=ES-DO style='mso-ansi-language: ES-DO'>Extracción del ADN</span></i></p>    ]]></body>
<body><![CDATA[<p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Método  <span class=spelle><i style='mso-bidi-font-style:normal'>Gaillard</i></span> y  <i style='mso-bidi-font-style:normal'>Strauss</i><sup>15</sup></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Se  maceraron los mosquitos de forma individual en nitrógeno líquido y se adicionaron  500 &#956;L del tampón de extracción (20 % SDS; 1 M <span class=spelle>Tris</span>-<span class=spelle>HCl</span> <span class=spelle>pH</span> 8,25; 0,5 M EDTA; 1 M sacarosa).  El homogenizado se incubó a 65 <span class=spelle><span style='font-family:Arial'>°</span>C</span> por 10 <span class=spelle>min</span>,  se añadieron 120 &#956;L de acetato de potasio 5 M, se puso en hielo por 10 <span class=spelle>min</span> y se centrifugó a 10 000 <i style='mso-bidi-font-style: normal'>g</i> por 10 min. El <span class=spelle>sobrenadante</span> se recogió,  se centrifugó nuevamente, se le adicionaron 35 &#956;L de mezcla de acetato (acetato  de sodio 4 M; 0,25 % <span class=spelle>acrilamida</span>) y 1 200 &#956;L de  etanol absoluto, incubándose 10 <span class=spelle>min</span> a temperatura ambiente.  El homogenizado se centrifugó a 10 000 <i style='mso-bidi-font-style:normal'>g</i>  por 20 <span class=spelle>min</span>, el precipitado obtenido se lavó con etanol  70 %, se centrifugó a 10 000 <i style='mso-bidi-font-style:normal'>g</i> por 10  <span class=spelle>min</span>, se secó a temperatura ambiente y se resuspendió  en 15 &#956;L del tampón <span class=spelle>Tris</span>&#8211;EDTA (TE 1x) (1  <span class=spelle>mM</span> <span class=spelle>Tris</span>-<span class=spelle>HCl</span>  <span class=spelle>pH</span> 8,0; 0,1 <span class=spelle>mM</span> EDTA <span class=spelle>pH</span> 8,0). Posteriormente, se adicionaron 2 &#956;L de <span class=spelle>RNAsa</span> (10 <span class=spelle>mg</span>/mL), se incubó 1 h  a 37 <span class=spelle><span style='font-family:Arial'>°</span>C</span>, se añadió  igual volumen de la mezcla <span class=spelle>cloroformo</span><span class=grame>:alcohol</span> <span class=spelle>isoamílico</span> (24:1) y se centrifugó  a 10 000 <i style='mso-bidi-font-style:normal'>g</i> por 10 min. El <span class=spelle>sobrenadante</span> se conservó a 4 <span class=spelle><span style='font-family:Arial'>°</span>C</span>.</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><i><span style='mso-bidi-font-size:11.0pt'>RAPD</span></i></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>La  amplificación del ADN se llevó a cabo en un volumen final de 25 &#956;L, que contenía  2,5 U <span class=spelle>Taq</span> ADN polimerasa (<span class=spelle>Promega</span>, USA), 2,5 &#956;L 10X tampón de reacción (100 <span class=spelle>mM</span> <span class=spelle>Tris</span>-<span class=spelle>HCl</span>;  <span class=spelle>pH</span> 8.3; 15 <span class=spelle>mM</span> <span class=spelle>MgCl<sub>2</sub></span>; 500 <span class=spelle>mM</span> <span class=spelle>KCl</span>; 0,01 % gelatina; <span class=spelle>Promega</span>, USA),  200 <span class=spelle>µM</span> de cada <span class=spelle>deoxinucleótidos</span>  (<span class=spelle>dNTP</span>) (<span class=spelle>Promega</span>, USA), 5 <span class=spelle>pmol</span> del cebador (OPA 7, <span class=spelle>Operon</span>  Technologies, California, USA) y 100 <span class=spelle>ng</span> de ADN molde.  </p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Se  utilizó un control negativo para cada ensayo donde la mezcla de reacción contenía  agua destilada estéril en lugar de ADN molde de <i style='mso-bidi-font-style: normal'>Aedes <span class=spelle>aegypti</span></i>, con el objetivo de descartar  cualquier contaminación. La reacción fue realizada siguiendo los perfiles de amplificación:  desnaturalización inicial a 94 <span class=spelle>°C</span> por 5 <span class=spelle>min</span>,  seguido por 40 ciclos de 1 <span class=spelle>min</span> a 94 <span class=spelle><span style='font-family:Arial'>°</span>C</span>,  1 <span class=spelle>min</span> a 35 <span class=spelle><span style='font-family: Arial'>°</span>C</span> y 2 <span class=spelle>min</span> a 72 <span class=spelle>°C</span>. En el ciclo final, el paso de extensión fue de 15 min.  El análisis de los fragmentos amplificados fue realizado en <span class=spelle>gel</span>  de <span class=spelle>agarosa</span> 1,4 % en <i style='mso-bidi-font-style: normal'>buffer</i> TBE 0,5X que contenía bromuro de <span class=spelle>etidio</span>  (0,5 <span class=spelle>mg</span>/mL) y visualizados con luz UV en un <span class=spelle>transiluminador</span>.</p><h4>Resultados</h4>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Los  valores dosis-respuesta y los factores de resistencia de las cepas Santiago de  Cuba y Santiago de Cuba Techo frente a 7 insecticidas son reportados en la tabla  1. Los mosquitos de la población de Santiago de Cuba presentan altos niveles de  resistencia al insecticida <span class=spelle>organofosforado</span> <span class=spelle>clorpirifos</span>  (16,91), esta población muestra además, moderados niveles de resistencia a otros  <span class=spelle>organofosforados</span> como: <span class=spelle>pirimifos</span>-<span class=spelle>metil</span>  (8,10), <span class=spelle>temefos</span> (5,59) y <span class=spelle>fentión</span>  (5,41), para el <span class=spelle>piretroide</span> <span class=spelle>cipermetrina</span>  también existieron moderados niveles de resistencia (7,83). Sin embargo en estos  estudios no se detectó resistencia a <span class=spelle>malatión</span> (1,77),  ni a la <span class=spelle>deltametrina</span> (4,75). En cambio, Santiago de  Cuba techo mostró una elevada resistencia al <span class=spelle>piretroide</span>  <span class=spelle>deltametrina</span> (114,38), alta resistencia a <span class=spelle>clorpirifos</span> (21,32), <span class=spelle>pirimifos</span>-<span class=spelle>metil</span> (10) y a la <span class=spelle>cipermetrina</span> (12,33),  se indica además moderada resistencia a 2 <span class=spelle>organofosforados</span>:  <span class=spelle>temefos</span> (5,51) y <span class=spelle>fentión</span> (5,41).  Finalmente no se detectó resistencia a <span class=spelle>malatión</span> (1,22),  similar a lo ocurrido en la otra cepa. </p>    <p class=MsoNormal align=center style='mso-margin-top-alt:auto;mso-margin-bottom-alt: auto;text-align:center'><b style='mso-bidi-font-weight:normal'>Tabla 1</b>. Se  observó, que este mecanismo se involucra también en la resistencia a <span class=spelle>temefos</span>  en la cepa de Santiago de Cuba (<span class=spelle>FS=</span> 6,45) (tabla 2). Finalmente, la frecuencia de la <span class=spelle>acetilcolinesterasa</span>  alterada fue muy baja en ambas poblaciones. </p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Se  empleó la electroforesis en <span class=spelle>geles</span> de <span class=spelle>poliacrilamida</span> para determinar los patrones de <span class=spelle>esterasas</span> elevadas en las poblaciones en estudio. Las bandas  de <span class=spelle>esterasas</span> fueron nombradas A ó B, de acuerdo con  la especificidad de la reacción con el reactivo <span style='font-family:Symbol;mso-ascii-font-family:"Times New Roman";mso-hansi-font-family: "Times New Roman";mso-char-type:symbol;mso-symbol-font-family:Symbol'><span style='mso-char-type:symbol;mso-symbol-font-family:Symbol'>a</span></span> o <span style='font-family:Symbol;mso-ascii-font-family:"Times New Roman";mso-hansi-font-family: "Times New Roman";mso-char-type:symbol;mso-symbol-font-family:Symbol'><span style='mso-char-type:symbol;mso-symbol-font-family:Symbol'>b</span></span> <span class=spelle>naftil</span> acetato. Estas <span class=spelle>esterasas</span>  fueron numeradas teniendo en cuenta la movilidad relativa, la cual fue calculada  como la distancia del origen a la banda correspondiente, divida por la distancia  total de la banda que más migró en 45 <span class=spelle>min</span> (<span class=spelle>Rm</span>).  El análisis de los patrones <span class=spelle>electroforéticos</span> revela  la elevación de la <span class=spelle>esterasa</span> A4 con un <span class=spelle>Rm</span> de 0,78. La intensidad de la banda de la <span class=spelle>esterasa</span> A4 en ambas cepas de Santiago de Cuba fue muy similar,  no así en la cepa de referencia <span class=spelle>Rockefeller</span>, que no  posee la elevación de esta (fig. 1). </p>    ]]></body>

Los mosquitos pertenecientes a la población de Santiago de Cuba  Techo presentan los mismos parches que los de la cepa de Santiago de Cuba.</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Se  utilizó la técnica del ADN polimórfico amplificado a la azar con el cebador OPA  7 para observar la variabilidad genética entre las 2 poblaciones analizadas, los  resultados mostrados en la figura 2 revelan el polimorfismo genético existente  entre las poblaciones en estudio y la poca variabilidad genética dentro de cada  cepa.</p>    <p align=center style='text-align:center'><b><span style='font-size:8.0pt; font-family:Verdana;color:#000099'><img width=427 height=339 id="_x0000_i1027" src=f0209205.jpg border=0></span></b></p>    <p align=center style='text-align:center'><b>Fig  2</b>. Patrones obtenidos por RAPD de mosquitos <i>Aedes aegypti </i>(línea 1:  marcador de peso molecular 1 Kb, Promega, USA), (línea 2: control negativo), (líneas  3, 4, 5: Rockefeller), (líneas 6-11: Santiago de Cuba), (líneas 12-16: Santiago  de Cuba Techo).</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>&nbsp;</p><h4><span style='mso-bidi-font-size:11.0pt'>Discusión</span></h4>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Los  resultados presentes en este trabajo son el primer reporte de la asociación que  existe entre la variabilidad genética, el cambio de conducta de reposo y la resistencia  a insecticidas en poblaciones de mosquitos <i style='mso-bidi-font-style: normal'>Aedes <span class=spelle>aegypti</span></i> de Cuba. La variación genética  encontrada entre las 2 cepas analizadas es similar a lo descrito por otros autores  en otras poblaciones de esta especie y de otras especies del género <i style='mso-bidi-font-style:normal'>Aedes</i>.<sup>16<span class=grame>,17</span></sup> Estudios de variabilidad genética en el nivel molecular  han demostrado que diferentes regiones del genoma poseen diferentes niveles de  cambio. Los genes de simple copia tienen un nivel relativamente bajo de sustitución,  porque cualquier cambio en la región codificadora puede producir serios defectos  en la codificación de la proteína. Efectos similares pueden ocurrir en regiones  no codificadoras del genoma. En cambio, el ADN repetitivo no está bajo tales restricciones  funcionales y la variabilidad en esas regiones del genoma es mayor que los genes  de simple copia.<sup>18</sup> Cualquier mutación que modifica el comportamiento  o la fisiología del insecto donde la secuencia blanco permanezca parcialmente  funcional puede inducir resistencia.<sup>19</sup></p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Cuando  los patrones del RAPD son utilizados para analizar las variaciones genéticas entre  las poblaciones, generalmente se encuentran altos niveles de polimorfismo.<sup>6,20,21</sup>  Los fragmentos de ADN obtenidos por la técnica del RAPD de las poblaciones analizadas  indicaron variación genética entre las cepas de Santiago de Cuba, se observó además  poco polimorfismo entre una misma población (fig. 2). La poca variabilidad detectada  en estos estudios por análisis morfológico y en <span class=spelle>geles</span>  de electroforesis de proteína no parece estar asociada con la resistencia a insecticidas,  ni con el cambio de conducta de reposo de los mosquitos implicados. Reportes de  otros autores plantean que bandas de patrones <span class=spelle>electroforéticos</span>  iguales muestran diferentes espectros de resistencia.<sup>22</sup> Sin embargo,  en el presente trabajo se muestra una estrecha relación entre las variaciones  genéticas de las cepas con la diferencia en los patrones de resistencia al <span class=spelle>piretroide</span> <span class=spelle>deltametrina</span> y con la  diferencia de conducta de reposo. </p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'><span style="mso-spacerun: yes">&nbsp;</span>Las 2 cepas analizadas presentan diferentes  niveles de resistencia al <span class=spelle>piretroide</span> <span class=spelle>deltametrina</span>. Existen varios reportes de resistencia a <span class=spelle>piretroides</span> en <i style='mso-bidi-font-style:normal'>Aedes  <span class=spelle>aegypti</span></i>.<sup>23-25</sup> Altos niveles de la enzima <span class=spelle>glutatión</span> <span class=spelle>transferasa</span> se han relacionado  con la resistencia a <span class=spelle>piretroides</span>.<sup>26,27</sup> El  incremento de la resistencia a <span class=spelle>deltametrina</span> en la cepa  de Santiago de Cuba Techo fue asociado con un incremento en la frecuencia de genes  de la GST, similares resultados en poblaciones de <i style='mso-bidi-font-style: normal'>Aedes <span class=spelle>aegypti</span> </i>de Santiago de Cuba fueron  reportados por <i>Rodríguez</i> y otros.<sup>1,28</sup> Estos apuntes coinciden  con los resultados obtenidos en este trabajo. A pesar de ser el <span class=spelle>malatión</span> un insecticida usado por mucho tiempo en Cuba para  el control del vector del dengue, no se presenta resistencia en estas poblaciones,  esto se atribuye a que se ha dejado de manejar en los programas de control. La  resistencia a <span class=spelle>cipermetrina</span> se explica por la aplicación  de este insecticida en sustitución del <span class=spelle>organofosforado</span>  <span class=spelle>malatión</span>, a partir del año 1986 en la campaña contra  el <i style='mso-bidi-font-style:normal'>Aedes <span class=spelle>aegypti</span></i>.  La resistencia a este insecticida no mostró grandes variaciones entre las cepas  en estudio. Los <span class=spelle>organofosforados</span> <span class=spelle>temefos</span>,  <span class=spelle>fentión</span>, <span class=spelle>pirimifos</span>-<span class=spelle>metil</span> y <span class=spelle>clorpirifos</span> sufren poca  variación en los factores de resistencia entre ambas cepas, resultado que puede  argumentarse por el empleo de estos como <span class=spelle>larvicidas</span>,  llegando a convertirse en presión de selección para las poblaciones de Santiago  de Cuba. </p>    ]]></body>
<body><![CDATA[<p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>Poder  entender la diferencia de conducta de reposo en las poblaciones de Santiago de  Cuba sería un aspecto ecológico novedoso e importante. Su conocimiento resultaría  de vital importancia en la confección de estrategias de control de los mosquitos  <i style='mso-bidi-font-style:normal'>Aedes <span class=spelle>aegypti</span></i>,  teniendo en cuenta los patrones de resistencia y los estudios relacionados en  la epidemiología del vector del dengue.</p>    <p class=MsoNormal style='mso-margin-top-alt:auto;mso-margin-bottom-alt:auto'>&nbsp;</p><h2><span lang=EN-GB style='mso-ansi-language:EN-GB'>Comparison  of 2 populations of <i>Aedes aegypti</i> mosquitoes from Santiago de Cuba with  different rest conduct <o:p></o:p></span></h2><h4><span lang=EN-GB style='mso-ansi-language:EN-GB'>Summary<o:p></o:p></span></h4>    <p><span lang=EN-GB style='mso-ansi-language:EN-GB'>Two  populations of <i>Aedes aegypti</i> that were collected in Santiago de Cuba during  the epidemics of 1971 were separated for having different rest habits, some of  them rested naturally on the walls up to 1 m high (Santiago de Cuba strain) and  the others were found resting in the roofs of the houses (Santiago de Cuba Techo  strain). These strains did not show significant differences as regards their morphological  characteristics. The mosquitoes corresponding to Santiago de Cuba Techo strain  presented the same patches that those of Santiago de Cuba. The resistance to organophosphate  insecticides is very similar in both populations; however, the Santiago de Cuba  Techo strain shows a higher resistance to pyrethroid deltamethrin than the Santiago  de Cuba strain. From the biochemical point of view and by using the DEF synergist,  it was proved that esterases are associated with the high resistance to clorpirifos  in both strains. It was not so with the MFO, which was demonstrated by means of  the piperomyl butoxide sinergist. Nevertheless, the GST enzyme seems to be responsible  for the high resistance to deltamethrin detected in the Santiago de Cuba Techo  strain due to the elevated frequency value of that gene in this strain. The random  amplified polymorphic DNA technique was used to observe the genetic variability  between the 2 populations.
