Objetivo: detectar la posible presencia de ciertos grupos de metabolitos secundarios mediante análisis fitoquímico preliminar y evaluar la actividad antioxidante, antiinflamatoria y antiproliferativa del extracto etanólico de corteza de Zanthoxylum fagara (L.) Sarg. (Rutaceae). Métodos: el extracto de corteza se sometió a un análisis fitoquímico preliminar, evaluándose mediante ensayos de actividad antioxidante (DPPH, TBARS y peroxidación inducida con FeSO4), antiinflamatoria (edema de oreja de ratón inducido con TPA) y antiproliferativa (se trataron células tumorales, y se usó proteína sulforrodamina B [SRB] en ensayo de microcultivo, se midió viabilidad y crecimiento celular). Resultados: en el extracto se detectó la presencia de alcaloides, flavonoides, taninos y terpenos, principalmente. La actividad antioxidante, a concentraciones de 10, 100 y 1 000 ppm, exhibió porcentajes de inhibición de 14,9; 84,5 y 92,5 %, respectivamente (frente al DPPH); de 92,8 % (a 1 000 ppm) (prueba TBARS), y de 4,2; 47,1 y 93,9 %, respectivamente, en la peroxidación inducida con FeSO4. La actividad antiinflamatoria mostró 45,3 %, y en el caso de la antiproliferación fue moderadamente activo contra las líneas celulares cancerosas K562, con valor de 55,5 %. En otras líneas evaluadas (U251, PC-3, HCT-15, MCF-7, y SKUL) el porcentaje fue inferior a 50 %. Conclusiones: se evidenció la presencia de alcaloides, flavonoides, taninos y terpenos. La capacidad antioxidante y antiinflamatoria fue representativa a concentraciones superiores a 100 ppm, mientras que la actividad antiproliferativa fue moderadamente efectiva contra líneas de células cancerosas leucémicas K562.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[Introduction: the Z. fagara species having multiple ethnobotanical uses, and a variety of secondary metabolites becomes a promising object of study at phytochemical and/or pharmacological settings. Objectives: to detect the possible presence of certain groups of secondary metabolites by means of a preliminary phytochemical analysis and to evaluate the antioxidant, anti-inflammatory and antiproliferative activities of the ethanolic extract from Zanthoxylum fagara (L.) Sarg. (Rutaceae)bark. Methods: the bark extract underwent preliminary phytochemical analysis using tests to evaluate antioxidant action (DPPH, TBARS test and FeSO4-induced peroxidation), anti-inflammatory activity (using mouse ear edema induced by acetate 12-O-tetradecanoylphorbol, TPA) and antiproliferative property (in treating tumor cells, sulphorodamine B protein (SRB) in microculture assay was used in addition to measuring cell viability and growth). Results: alkaloids, flavonoids, tannins and terpenes were found in the extract. The antioxidant activity at 10, 100 and 1000 ppm concentrations, exhibited inhibition percentages of 14.9; 84.5 and 92.5 % respectively [compared to DPPH]; 92.8 % (1 000 ppm) [TBARS test], and 4.2; 47.1; and 93.9 %, respectively, in the FeSO4-induced peroxidation. The anti-inflammatory activity showed inhibition percentage of 45.3 %, and in the case of anti-proliferation, the extract was moderately active against cancer cell lines K562, being the inhibition percentage equals to 55.5 %. In the other tested lines (U251, PC-3, HCT-15, MCF-7, and Skule) the inhibition percentage was lower than 50 %. Antioxidant and anti-inflammatory    properties were observed at above 100 ppm extract concentrations, whereas the    antiproliferative activity was moderately effective against leukemia cancer    cell lines K562. </font>      <P>      <P><font face="Verdana" size="2"><B>Key words</B>: Phytochemical analysis, antioxidant    action, antiproliferative activity, anti-inflammatory activity, <I>Zanthoxylum</I>,    Rutaceae. </font> <hr size="1" noshade>     <p>&nbsp;</p>     <p>&nbsp;</p>     <P>      <P><font face="Verdana" size="3"><B>INTRODUCCI&Oacute;N</B> </font>      <P>      <P><font face="Verdana" size="2">El g&eacute;nero <I>Zanthoxylum</I> presenta    un h&aacute;bitat como arbustos o arbolillo caducifolio, de 4 a 6 m de altura    como m&aacute;ximo, de tallos espinosos y ramillas de color marr&oacute;n.<SUP>1</SUP>    A nivel mundial, el g&eacute;nero <I>Zanthoxylum</I> se encuentra distribuido    en el continente americano y asi&aacute;tico; mantiene una elevada distribuci&oacute;n    en Colombia, con reportes de su presencia en 23 de los 32 departamentos que    conforman su divisi&oacute;n pol&iacute;tica.<SUP>2</SUP> </font>     <P><font face="Verdana" size="2">En la medicina folcl&oacute;rica el g&eacute;nero    <I>Zanthoxylum</I> es muy usado, se citan algunos ejemplos: <I>Zanthoxylum xanthoxyloide</I>,    que reporta usos contra la tos, fiebre, dolor de muela, mordedura de serpiente,    enteritis, disenter&iacute;a, diarrea, uretritis y contra la estomatitis;<SUP>3,4</SUP>    <I>Zanthoxylum chalybeum, </I>con usos<I> </I>contra la malaria, resfriados,    tos, dolor de muela, heridas, resfriados y neumon&iacute;a;<SUP>5</SUP> <I>Zanthoxylum    usambarense </I>(tratamiento del reumatismo);<SUP>5</SUP> <I>Zanthoxylum tetraspermun    </I>contra la diarrea, reumatismo y dispepsia;<SUP>6,7</SUP> <I>Zanthoxylum    integrifoliolum</I> que se utiliza como remedio en la mordedura de serpiente,    dispepsia y t&oacute;nico contra la fiebre;<SUP>8</SUP> <I>Zanthoxylum hyemale    </I>como antiespasm&oacute;dico, astringente y agente t&oacute;nico;<SUP>9</SUP>    y <I>Zanthoxylum liebmannianum</I> frecuentemente usado contra el dolor de est&oacute;mago,    amebiasis, anest&eacute;sico local y par&aacute;sitos de tipo intestinal.<SUP>6,10</SUP>    Algunos ejemplos de actividades biol&oacute;gicas comprobadas pueden ser mencionadas,    las cuales se soportan por el gran uso que tienen estos en la medicina folkl&oacute;rica,    a saber: <I>Zanthoxylum xanthoxyloides</I> (actividad antimicrobial);<SUP>3,4,11</SUP>    <I>Zanthoxylum budrunga </I>(citot&oacute;xica);<SUP>12,13</SUP> <I>Zanthoxylum    integrifoliolum</I> (antiagregaci&oacute;n plaquetaria);<SUP>6</SUP> <I>Zanthoxylum    americanum</I> (actividad antitumoral y de letalidad hacia la <I>Artemia salina</I>);<SUP>14</SUP>    <I>Zanthoxylum syncarpum</I> (antiplasmodial);<SUP>7</SUP> <I>Zanthoxylum simulans</I>    (antiagregaci&oacute;n plaquetaria);<SUP>15</SUP> <I>Zanthoxylum chiloperone</I>    (actividad leishmanicida);<SUP>16</SUP> <I>Zanthoxylum liebmannianum</I> (amebicida);<SUP>10,17</SUP>    y <I>Zanthoxylum chalibeam</I> (actividad antibacterial y antiinflamatoria).<SUP>18,19</SUP>    </font>     ]]></body>
<body><![CDATA[<P><font face="Verdana" size="2">Los metabolitos secundarios m&aacute;s representativos    del g&eacute;nero <I>Zanthoxylum </I>son alcaloides, lignanos, amidas, cumarinas,    terpenos, flavonoides, pero tambi&eacute;n pueden hallarse esteroles y cromonas.<SUP>20</SUP>    De la especie <I>Z. fagara</I> se han encontrado metabolitos como: sinefrina,<SUP>21</SUP>    meridinol,<SUP>22</SUP> escopoletina y skimmianina.<SUP>23</SUP> As&iacute;    mismo, en su aceite esencial se ha encontrado citronellol, <I>cis</I>-3-hexenol,    y el citronellal, como metabolitos mayoritarios entre otros.<SUP>24</SUP> De    <I>Z. fagara</I> se reporta actividad antif&uacute;ngica contra los microorganismos    <I>Saccharomyces cerevisiae</I>, <I>Microsporum canis</I> y <I>Trichophyton    mentagrophytes</I>.<SUP>25</SUP> </font>      <P>     <P><font face="Verdana" size="2"><B><font size="3">M&Eacute;TODOS</font></B> </font>      <P><font face="Verdana" size="2"><I>Material vegetal</I> </font>     <P><font face="Verdana" size="2">La muestra de la especie <I>Zanthoxylum fagara</I>    (L.) Sarg. (Rutaceae) fue recolectada en los Montes de Mar&iacute;a, del Departamento    de Bol&iacute;var, por <I>Giovanni Montes Arrieta</I> y <I>Olimpo Garc&iacute;a</I>.    Una muestra de herbario se encuentra en el Herbario de la Universidad del Magdalena    bajo la codificaci&oacute;n: &quot;<I>Zanthoxylum fagara</I> (L.) Sarg. Colombia.    Bol&iacute;var. Montes de Mar&iacute;a, marzo de 2007.Y Montes y O. Garc&iacute;a    No 04 (UTMC)&quot;. </font>     <P>      <P><font face="Verdana" size="2"><I>Obtenci&oacute;n del extracto</I> </font>     <P><font face="Verdana" size="2">La corteza seca y molida (700 g) se someti&oacute;    a extracci&oacute;n por percolaci&oacute;n con etanol a 96 %; el extracto fue    concentrado por remoci&oacute;n del solvente en un rotaevaporador Laborata 4000    (Heidolph) a presi&oacute;n reducida; se obtuvieron 28 g de extracto. </font>      <P>     <P><font face="Verdana" size="2"><I>Ensayo fitoqu&iacute;mico preliminar</I> </font>     ]]></body>
<body><![CDATA[<P><font face="Verdana" size="2">1,02 g del extracto etan&oacute;lico de la corteza    de <I>Zanthoxylum fagara</I> se utiliz&oacute; para la determinaci&oacute;n    fitoqu&iacute;mica preliminar de acuerdo con lo descrito por <I>Siddiqui</I>    y otros.<SUP>26</SUP> </font>     <P>      <P><font face="Verdana" size="2"><I>Ensayo de actividad antioxidante</I> </font>     <P><font face="Verdana" size="2">Para la reducci&oacute;n del radical libre 2,2-difenil-1-picrilhidracilo    (DPPH) se siguieron los protocolos propuestos por <I>G&aacute;mez</I><SUP>27</SUP>    y <I>Cavin</I>,<SUP>28</SUP> donde se tomaron 50 <font face="Symbol">m</font>L    de extracto etan&oacute;lico de corteza de <I>Zanthoxylum fagara</I> y se ensayaron    a concentraciones de 10, 100 y 1 000 ppm. <P>     <P>     <P>     <P>      <P>      <P><font face="Verdana" size="3"><B>DISCUSI&Oacute;N</B> </font>      <P><font face="Verdana" size="2">El an&aacute;lisis fitoqu&iacute;mico preliminar    del extracto etan&oacute;lico de la corteza de <I>Z. fagara</I> evidenci&oacute;    la posible presencia de alcaloides, lo que es muy com&uacute;n en especies del    g&eacute;nero <I>Zanthoxylum</I>. Tambi&eacute;n se encontraron resultados positivos    para flavonoides (que mostraron una tonalidad violeta, propia de la <font face="Symbol">n</font><font size="2">-</font>pirona),    taninos (que se evidenciaron por la coloraci&oacute;n de un precipitado azul)    y terpenos (lactonas terp&eacute;nicas), por la tonalidad rojiza y azul-verdosa    observada. La actividad antioxidante concuerda con la presencia de metabolitos    detectada, en la que el antioxidante puede donar un electr&oacute;n al radical    peroxilo o a cualquier otra especie oxidante, formando entre los productos un    cati&oacute;n radical del antioxidante (ArO&#149;+).<SUP>33</SUP> Dicha actividad    presenta valores comprendidos entre 15 y 93 % (indicando que la actividad es    dependiente de la dosis utilizada) para el ensayo DPPH; 93 % y 4-94 % para el    ensayo TBARS y para la peroxidaci&oacute;n inducida repectivamente; se encuentrra    una muy buena correlaci&oacute;n entre los 3 ensayos con diferencias absolutas    menores que 2 %. Esta actividad puede deberse a la presencia de algunos metabolitos    de tipo flavonoide, que al reaccionar con el radical (por ejemplo el DPPH),    el grupo hidroxi del anillo arom&aacute;tico del flavonoide estabiliza el radical    resultante por donaci&oacute;n de electrones y aumenta por tanto la capacidad    antioxidante.<SUP>34</SUP> Dentro de estos flavonoides se pueden citar hiperina,<SUP>35,10</SUP>    quercetrina,<SUP>36</SUP> 3,5-diacetiltambulina,<SUP>37</SUP> (2S)-naringenina,<SUP>38</SUP>    quercetina, foeniculina, isorhamnetina<SUP> </SUP>o rutina,<SUP>35</SUP> entre    otros flavonoides comunes en el g&eacute;nero, los cuales han presentado buena    actividad antioxidante. No obstante, la capacidad antioxidante puede deberse    tambi&eacute;n a la presencia de lignanos, como por ejemplo la cubebina; o lignanos    tipo butirolactonas<SUP>39,40</SUP> e incluso alcaloides, especialmente del    tipo quinol&iacute;nico como la schinifolina.<SUP>41</SUP> </font>      <P><font face="Verdana" size="2">En la actividad biol&oacute;gica antiinflamatoria,    se mostro una acci&oacute;n moderadamente efectiva contra la inflamaci&oacute;n    con un valor promedio de 45,3 % de inhibici&oacute;n. Una v&iacute;a de acci&oacute;n    podr&iacute;a ser mediante la inhibici&oacute;n de la bios&iacute;ntesis de    prostaglandinas y leucotrienos, que involucra la acci&oacute;n de las ciclooxigenasas,<SUP>42-45</SUP>    lo cual podr&iacute;a ser confirmado en ensayos de actividad de inhibici&oacute;n    enzim&aacute;tica. La actividad antiinflamatoria mostr&oacute; un valor comparable    al reportado en la literatura (77,7 %), cuando se utiliz&oacute; en una dosis    doble.<SUP>46</SUP><B> </B> </font>     <P><font face="Verdana" size="2">En la actividad biol&oacute;gica antiproliferativa,    el extracto result&oacute; razonablemente eficiente a la concentraci&oacute;n    utilizada (20 <font face="Symbol">m</font>g/mL) en la inhibici&oacute;n del    crecimiento de c&eacute;lulas de leucemia K562 con porcentaje de inhibici&oacute;n    de 55,5 %, que mostr&oacute; una selectividad marcada, puesto que para otras    l&iacute;neas celulares se obtuvo un porcentaje de inhibici&oacute;n inferior    a 50 %, en un intervalo de 15 a 36 %. Este resultado abre la posibilidad dirigida    al uso potencial de este extracto en estudios posteriores en casos de leucemia,    con la finalidaf de encontrar los principios activos responsables de la actividad.    </font>     ]]></body>
