Las familias Moraxellaceae y Helicobacteraceae han sido ampliamente estudiadas, porque comprenden agentes patógenos causantes de numerosas enfermedades en humanos, pero pocas investigaciones han estado dirigidas a la identificación de las características moleculares que puedan distinguir a sus miembros de otros grupos de bacterias; de manera que los presentes resultados constituyen un aporte al conocimiento de la genética y la bioquímica de estas familias y proveen herramientas moleculares para la clasificación taxonómica y el diagnóstico de especies patógenas]]></p></abstract>

<body><![CDATA[<p>&nbsp;</p>     <p>&nbsp;</p>         <P><font size="3" face="Verdana, Arial, Helvetica, sans-serif"><B>INTRODUCCI&Oacute;N</B></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">La familia <I>Moraxellaceae</I>, incluida en la clase <I>Gammaproteobacteria</I>, comprende a los g&eacute;neros <I>Moraxella</I>, <I>Acinetobacter</I> y <I>Psychrobacter.</I> Algunas especies de <I>Moraxella </I>son importantes en el contexto de la medicina, por ser par&aacute;sitas de las membranas mucosas        del hombre y de otros mam&iacute;feros.  Asimismo la especie <I>Psychrobacter immobilis</I> y varias cepas de <I>Acinetobacter</I> han sido registradas como agentes pat&oacute;genos oportunistas, causantes        de numerosas infecciones        intrahospitalarias.<SUP>1-4</SUP></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">En la clase <I>Epsilonproteobacteria</I> se encuentra la familia <I>Helicobacteraceae</I>, que incluye el g&eacute;nero <I>Helicobacter</I>.<SUP>5</SUP> La bacteria <I>Helicobacter        pylori</I> es el agente causal reconocido de        varias enfermedades gastrointestinales en el hombre, como la gastritis cr&oacute;nica, las &uacute;lceras        p&eacute;pticas, el adenocarcinoma de est&oacute;mago y el linfoma de tejido linfoide asociado a la mucosa        g&aacute;strica.<SUP>6</SUP></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">A pesar de que las familias <I>Moraxellaceae</I> y <I>Helicobacteraceae</I> han sido        ampliamente estudiadas, pocas investigaciones han estado dirigidas a la determinaci&oacute;n de        marcadores moleculares para el diagn&oacute;stico de las especies pat&oacute;genas que estas contienen.</font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">La disponibilidad de genomas y proteomas completamente secuenciados para un        gran n&uacute;mero de bacterias, constituye una oportunidad para la taxonom&iacute;a y el        diagn&oacute;stico molecular.  De acuerdo con la metodolog&iacute;a propuesta por Gupta, la comparaci&oacute;n        de secuencias de prote&iacute;nas de diferentes especies mediante el alineamiento m&uacute;ltiple,        permite hallar marcadores moleculares que pueden ser empleados en la identificaci&oacute;n de        bacterias.<SUP>7</SUP></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">Teniendo en cuenta que ambas familias comprenden especies de importancia en el    &aacute;mbito m&eacute;dico, el presente estudio se bas&oacute; en la mencionada metodolog&iacute;a para la determinaci&oacute;n        de inserciones en prote&iacute;nas relacionadas con el metabolismo del &aacute;cido        desoxirribonucleico (ADN), que constituyan marcadores moleculares en la taxonom&iacute;a y diagn&oacute;stico de        especies pat&oacute;genas. </font>     <p>&nbsp;</p>         <P><font size="3" face="Verdana, Arial, Helvetica, sans-serif"><B>M&Eacute;TODOS</B></font>     ]]></body>
<body><![CDATA[<P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">La investigaci&oacute;n se llev&oacute; a cabo en el Departamento de Biolog&iacute;a y Geograf&iacute;a de la        Universidad de Oriente en Santiago de Cuba y para ello fueron escogidas secuencias de las prote&iacute;nas        ADN polimerasa III (subunidad alfa) y ADN polimerasa I pertenecientes a especies del <I>phylum</I> <I>Proteobacteria</I>, obtenidas de bases de datos internacionales a partir del servidor        <em>National Center of Biotechnology Information</em>.  Posteriormente fueron comparadas mediante        el programa ClustalX2,<SUP>8</SUP> considerando los par&aacute;metros sugeridos por Hall: alineamiento par a        par (apertura de gap 35 y extensi&oacute;n de gap 0,75) y alineamiento m&uacute;ltiple (apertura de gap 15        y extensi&oacute;n de gap 0,30).<SUP>9</SUP></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">Los alineamientos obtenidos fueron analizados mediante una inspecci&oacute;n visual        para identificar las inserciones, de las cuales se consideraron relevantes aquellas que        estuvieran flanqueadas por regiones conservadas, como fue propuesto por        Gupta.<SUP>7</SUP></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">La lista de las especies incluidas con su nomenclatura correspondiente es mostrada en        el anexo.  En cada figura la nomenclatura es seguida por el n&uacute;mero de acceso de la        secuencia en la base de datos.  Los puntos muestran identidad con el amino&aacute;cido de la        primera secuencia en el alineamiento y los guiones (-) representan brechas que indican ausencia        de la inserci&oacute;n, que est&aacute; se&ntilde;alada por un cuadro. </font>     <p>&nbsp;</p>         <P><font size="3" face="Verdana, Arial, Helvetica, sans-serif"><B>RESULTADOS</B></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">La <a href="/img/revistas/san/v22n1/f0106221.gif">figura 1</a> muestra una inserci&oacute;n de 11 amino&aacute;cidos en la prote&iacute;na ADN polimerasa        I, caracter&iacute;stica de los g&eacute;neros <I>Psychrobacter </I>y<I> Moraxella</I>, pero ausente en el        g&eacute;nero <I>Acinetobacter</I> de la familia <I>Moraxellaceae</I>.</font>     
<P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">El alineamiento de la prote&iacute;na ADN polimerasa III (subunidad alfa) evidenci&oacute; una        inserci&oacute;n de un amino&aacute;cido exclusivo del g&eacute;nero <I>Acinetobacter </I>(<a href="/img/revistas/san/v22n1/f0206221.gif">figura 2</a>), adem&aacute;s de otra inserci&oacute;n        de 4 a 17, caracter&iacute;stica de algunas especies de <I>Helicobacter </I>(<a href="/img/revistas/san/v22n1/f0306221.gif">figura 3</a>).     </font>     
<p>&nbsp;</p>         <P><font size="3" face="Verdana, Arial, Helvetica, sans-serif"><B>DISCUSI&Oacute;N</B></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">La inserci&oacute;n compartida por los g&eacute;neros <I>Psychrobacter </I>y <I>Moraxella </I>est&aacute; bien conservada        en las secuencias de las especies de<I> Psychrobacter</I>, lo que permite diferenciarlas de las        especies de<I> Moraxella</I>.  Las especies pat&oacute;genas <I>M. catarrhalis </I>y <I>M. nonliquefaciens</I> han sido        aisladas en humanos a partir de la cavidad nasal y de muestras del tracto respiratorio de        pacientes con bronquitis cr&oacute;nica y otras infecciones de las v&iacute;as        respiratorias;<SUP>10</SUP> un n&uacute;mero        significativo de sus cepas producen beta-lactamasa, lo que ha condicionado su resurgimiento        como agentes pat&oacute;genos en este tipo de        infecciones.<SUP>2</SUP></font>     ]]></body>
<body><![CDATA[<P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">Igualmente la inserci&oacute;n exclusiva de un amino&aacute;cido en <I>Acinetobacter</I> en la prote&iacute;na ADN polimerasa III (subunidad alfa) resulta de gran valor para la taxonom&iacute;a e identificaci&oacute;n        a nivel de g&eacute;nero.  Entre las especies que la presentan est&aacute; el <I>A. baumanii</I>, que ha emergido como uno de los microorganismos hospitalarios de mayor relevancia a escala        mundial.<SUP>4</SUP> Existe una clara asociaci&oacute;n entre la infecci&oacute;n por estas bacterias y el uso de        instrumentales hospitalarios, y las infecciones resultan en septicemias, meningitis, endocarditis,        neumon&iacute;as e infecciones del tracto urinario.  Algunas cepas aisladas en Cuba han mostrado resistencia        a m&uacute;ltiples antibi&oacute;ticos, como la ampicilina, gentamicina, amikacina y        ciprofloxacina.<SUP>11</SUP></font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">Muchas especies de <I>Helicobacter </I>son pat&oacute;genas del hombre y est&aacute;n asociadas a        tres enfermedades, generalmente relacionadas con su nicho ecol&oacute;gico        natural.<SUP>12</SUP> Entre las especies que comparten la inserci&oacute;n de 5 amino&aacute;cidos en la prote&iacute;na ADN polimerasa        III (subunidad alfa) est&aacute; el <I>H.          pylori</I>, reconocida como la principal bacteria causante de    &uacute;lceras p&eacute;pticas, gastritis atr&oacute;fica y c&aacute;ncer        g&aacute;strico.<SUP>5,13</SUP> Es importante se&ntilde;alar que la        inserci&oacute;n permite una distinci&oacute;n de esta especie debido a que la secuencia de amino&aacute;cidos difiere en        la cuarta posici&oacute;n respecto a la <I>H.          cetorum</I>, que tambi&eacute;n la presenta.</font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">La inserci&oacute;n es de 17 amino&aacute;cidos en 3 de las especies de <I>Helicobacter</I>:<I> H. pametensis</I>, <I>H. typhlonius </I>y<I> H. hepaticus</I>; las dos &uacute;ltimas comparten las mismas secuencias de        amino&aacute;cidos, lo que resulta de gran valor desde el punto de vista m&eacute;dico, puesto que el <I>H. heilmannii </I>ha sido aislado de las muestras tomadas de pacientes con        gastroenteritis.<SUP>14</SUP>     </font>              <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">Hay que se&ntilde;alar que las especies ent&eacute;ricas de <I>Helicobacter </I>no son frecuentemente notificadas como agentes causales de enfermedades en humanos, pero su incidencia        pudiera estar significativamente subestimada, debido a los inadecuados procedimientos        de aislamiento e        identificaci&oacute;n.<SUP>13</SUP> Considerando las dificultades en la identificaci&oacute;n de        especies comprendidas en este g&eacute;nero y dada la importancia cl&iacute;nica de algunas de ellas,        la determinaci&oacute;n de marcadores moleculares espec&iacute;ficos de este importante grupo resultar&iacute;a        de gran relevancia para la medicina.</font>     <P><font size="2" face="Verdana, Arial, Helvetica, sans-serif">La metodolog&iacute;a desarrollada por Gupta ha sido utilizada por otros autores en el an&aacute;lisis        de diferentes grupos de bacterias.<SUP>15,16</SUP> Las inserciones halladas en este estudio constituyen        una herramienta molecular &uacute;til para el dise&ntilde;o de protocolos de diagn&oacute;stico que        permitir&aacute;n establecer futuras estrategias respecto a la epidemiolog&iacute;a y el tratamiento en        enfermedades ocasionadas por dichos agentes pat&oacute;genos.
