La investigación basada en plantas medicinales es una de las estrategias recomendadas para la búsqueda de nuevas alternativas terapéuticas contra la leishmaniasis. En este estudio, se demuestran las potencialidades de productos derivados de Bixa orellana L. sobre Leishmania amazonensis, agente causal de la leishmaniasis cutánea (LC).  Métodos.  Se obtuvieron los productos de las semillas de Bixa (extractos, fracciones, subfracciones y aceite esencial), los cuales fueron evaluados en diferentes modelos biológicos in vitro e in vivo.  Resultados.  El extracto hidroalcohólico causó inhibición del crecimiento de promastigotes y amastigotes, fue capaz de controlar el desarrollo de la LC experimental en ratones BALB/c (100 mg/kg/día/15 días/intraperitoneal), y se obtuvo su perfil químico. Posteriormente, se realizó un estudio bioguiado con 4 fracciones y 28 sub-fracciones, de las cuales BO-B1 y BO-B3 resultaron promisorias y se identificaron un grupo de isómeros alifáticos (bixina y la norbicina). El aceite esencial fue el otro producto obtenido, el cual fue caracterizado químicamente, inhibió el crecimiento de amastigotes, mostró una citotoxicidad 7 veces menor respecto a la actividad sobre el parásito, fue efectivo contra la LC experimental (30 mg/kg/día/15 días/intraperitoneal e intralesional) y se evidenció una inhibición de manera específica del consumo de oxígeno y el complejo II mitocondrial. El aceite esencial formulado en nanococleatos demostró una alta eficacia en el modelo murino al administrar 4 dosis por vía intralesional. Los resultados de esta investigación demuestran las potencialidades de las semillas de B. orellana sobre L. amazonensis para el tratamiento de la LC, enfermedad con diversas presentaciones clínicas que cuenta actualmente con limitadas opciones terapéuticas.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[ABSTRACT  Introduction.  Research based on medicinal plants is one of the recommended strategies for the search for new therapeutic alternatives against leishmaniasis. In this study, the potential of products derived from Bixa orellana L. on Leishmania amazonensis, a causal agent of cutaneous leishmaniasis (CL), is demonstrated.  Methods. Bixa seed products (extracts, fractions, sub-fractions and essential oil) were obtained, which were evaluated in different in vitro and in vivo biological models.  Results.  The hydroalcoholic extract caused inhibition of the growth of promastigotes and amastigotes, was able to control the development of experimental CL in BALB/c mice (100 mg/kg/day/15 days/intraperitoneal), and its chemical profile was obtained. Subsequently, a bioguided study was performed with 4 fractions and 28 sub-fractions, which BO-B1 and BO-B3 were promising and a group of aliphatic isomers (bixin and norbicin) were identified. The essential oil was the other product obtained, which was chemically characterized, inhibited the growth of amastigotes, showed a cytotoxicity seven times lower with respect to the activity on the parasite, was effective against the experimental CL (30 mg/kg/day/15 days/intraperitoneal and intralesional) and a specific inhibition of oxygen consumption and mitochondrial complex II was evidenced. The essential oil formulated in nanocochleates showed high efficacy in the murine model when 4 doses were administered intralesionally. The results of this research demonstrate the potential of B. orellana seeds on L. amazonensis for the treatment of CL, a disease with various clinical presentations that currently has limited therapeutic options.]]></p></abstract>
