Abstract

Stevia rebaudiana Bertoni is worldwide known for its medicinal properties. The objective of this work was to obtain genetic profiles of in vitro grown plants or propagated by cuttings to detect genetic polymorphisms. For this, two DNA extraction protocols were first evaluated, both with modifications. The purified samples were used for random amplification of polymorphic DNA with six pairs of primers. A matrix of data was made from the band patterns. The similarity and genetic distance between both groups of plants were calculated. Genomic DNA was obtained only when the modified protocol proposed by Doyle and Doyle was applied. One of the six primers used was effective for the amplification of DNA segments. In this work, we provide a fast, simple and efficient DNA purification protocol for S. rebaudiana leaf samples. This might be applied for the study of genetic variability and the detection of somaclonal variations using other PCR-based techniques.
