Abstract

The enzyme tyrosinase is related to the enzymatic darkening of fungi, and it is considered as the main cause of the decrease in the commercial value. The 4j3p A structure of Aspergillus oryzae was selected as template for his similitude with the sequence of the hypothetical tyrosinase from Pleurotus ostreatus PC15. The models were built using the Swiss-model, I-TASSER and Phyre2 servers, and validated by means of the Ramachandran diagram supplied by Procheck, the Verify3D and ProSA servers. Three 3D models were obtained for the hypothetical tyrosinase enzyme of Pleurotus ostreatus PC15, validated by different computational strategies. Tyr Swiss was the best model with 85,5% of the amino acid residues under the favored region, and a z-score of -7,15 in ProSA. The Verify 3D value of 80.73%, indicated that the environment of the model profile was appropriated. The obtained models may be useful for discovery new enzyme inhibitors with industrial applications.
