Abstract

Thirty-three ejaculations of white New Zealand rabbits were obtained to perform the separation of "Y" spermatozoa by different density gradients (10, 15 and 20%) of Human Serum Albumin (HSA). The characteristics evaluated in fresh semen were: volume, progressive motility percentage, live spermatozoa, normal morphology and concentration, being these values: 1.19 ml, 82.72%, 90.90%, 89.96% and 773.63X108 respectively. Semen was diluted 1:1 in a medium of sodium citrate and glucose; then, 1ml of diluted semen was added in tubes containing gradients of albumin at 20%, 15% and 10%, at a temperature of 37.5oC. Latter, each of the factions was retired within intervals of 10 minutes each, determining motility percentage, live spermatozoa and spermatic concentration in each fraction. An important decrease of fresh semen motility was observed (82.72%) compared to the progressive motility in the HSA layer at 20%, which was 35.45%. As spermatozoa reach the layers of higher density, the percentage of the live ones decreases being 88.39, 86.21 and 83.06 for the 10, 15 and 20% of HAS respectively, observing the same related to spermatic concentration (504.24, 376.06 and 211.81X106/ml). From 30 births after artificial insemination with spermatozoa obtained from the layer with 20% of HSA, 187 rabbits were born being the 72.72% males and 27.27 females. The results indicate that it is possible to perform the selection of "Y" spermatozoa in rabbit semen and consequently produce more males, by the use of density gradients of HSA.
