Cumulus-oocyte complexes (COCs) were extracted from buffalo ovaries (Bubalus bubalis) slaughtered in slaughterhouse. COCs were transported to the laboratory for six hours in 500 ml of saline solution at 29.1°C. COCs extraction was carried out between 2 and 8 mm of diameter using two methods, the follicular puncture (FP) with 18Gx1½ needle and surgical dissection (SD) of follicles using an ophthalmic micro-scalpel. In order to classify CCOs, the integrity of the cumulus layers and the oocytes cytoplasm characteristics were taken into account. The results showed significant differences of 99% in the average of COCs recovery and their quality when compared to FP and SD methods. By the FP method, the average of COCs recovery was of 28 and 2.7 COCs per ovary with quality obtained, which represents 20,43% of all oocytes recovered in this group. However, the SD method achieved an average of COCs recovery of 139 and 6.3 COCs per ovary, representing 59,4% of all oocytes recovered for this group. These results demonstrate that the SD method for COCs collection is more efficient than the FP method.
