PEREZ PEREZ, Jorge L et al. Effect of 2,4-diclorophenoxiacetic acid on embryogenic response of soybean cultivar INCASoy-27. cultrop [online]. 2013, vol.34, n.3, pp. 40-44. ISSN 0258-5936.
The aim of this work was to evaluate the effect of 2,4-dichlorophenoxiacetic acid (2,4-D) on the embryogenic response of Cuban soybean cultivar INCASoy-27. Immature zygotic cotyledons between 3,0-4,0 mm length were placed with the abaxial surface on the culture medium for embryos induction. The culture medium was formulated with concentrations of 15, 25, 35, 40 and 45 mg.L-1 2,4-D. After four weeks in culture medium, the number of embryogenic cotyledons in the total of cotyledons and number of somatic embryos by embryogenic cotyledon was quantified. The percentage of immature zygotic cotyledons that produced somatic embryos varied between 23-93 % with 2,4-D concentration in the culture medium. The average number of somatic embryos formed per immature cotyledon fluctuated from 1,2 to 7,9 while in embryogenic cotyledons was 5,3 to 8,5 somatic embryos. The major response of this morphogenetic route was obtained at a concentration range of 40 and 45 mg.L-1 2,4-D in the culture medium. A differentiation process of somatic embryos with asynchrony in morphology began in the fourth week of culture.
